Gen IIm assay(modified Beckman-Coulter 2nd-generation antimullerian hormone assay)を用いることによって室温で保存された検体においてより一定の結果が得られるか否か、また、オリジナルの Gen IIassayと比較し希釈によってもより一定の値が得られるか否か比較検討した。また、新たに得られたAMH値と胞状卵胞数との関係についても調査した。不妊治療のため検査を受けた21~ 46 歳の678名の連続した女性を対象にコホート研究を行った。すでに胞状卵胞数が知られている女性を対象にGen IIm assayを用いてAMHを測定した。さらに、AMHレベルをオリジナルのassay法と変法を用いたassay法で測定をしその値を比較した。
Gen IIm assayで測定した結果、希釈してもAMHの値はそれと比例し変化した。また、AMHレベルは7日間室温で検体を保存することによって平均12.1%の減少をみたが、オリジナルのGen II assayを用いた場合にはかなりの上昇が認められた。

Gen IIm assay で得られた値はGen II assay で得られた値よりも平均51.4%高い値を示した。Gen IIm とDiagnostic Systems Laboratories AMH assay で調べた歴史的データとの変換指数は 1.35であった。GenIIm assayのAMHの検査値と胞状卵胞数との間には直線的な関係が認められた。

新しく開発された自動AMHの測定装置であるElecsys AMH(Roche; Elecsys)とAccess AMH(BeckmanCoulter; Access)を用いた AMH の測定結果と従来から用いられている AMH Gen II ELISA(enzyme-linked immunosorbent assay; Gen II; Beckman Coulter)と AMH ELISA(Ansh Labs)assayの測定結果を比較した。
2014年 9~ 10月にかけてIVF前の83名の患者を対象に血中AMHの測定を行い、前方視的にその結果を分析した。測定内CVはいずれも低くAnshでは9.0%以下、Gen II では5.8%以下、Access では10.7%以下、Elecsysでは2.8%以下という結果が得られた。

甲状腺自己免疫(thyroid autoimmunity、TAI)は原発性卵巣不全の女性においてよく共存する内分泌疾患である。いくつかの研究においてTAIと卵巣予備能の低下との相関が示されている。しかし、卵巣予備能の低下をみた女性における TAI の発現率に関して確かな根拠は得られていない。そこで、TAI と卵巣予備能低下との相関について調べた。大学病院において後方視的データを用い大規模な横断面的研究を行った。
新たに市場に導入された完全自動AMH測定法はAMH Gen II assayと同じようにAMHを測定できるが、測定感度は有意に高いという結果が得られた。

標準化を欠いているために AMH ELISA assay の結果を比較することは時に難しいこともある。BCIAMHGen II assayはこの数年間、臨床の基準となる測定法として用いられてきた。AMH Gen II antibodycompositionを基にした2つの新たな高感度AMH免疫自動測定法が2014年9月から利用できるようになった。そこで、2つの異なった企業から市場に出された2つの新たな完全自動AMH assay法と現在臨床的に基準となっているAMH Gen II enzyme-linked immunosorbent assay (ELISA)と比較した。
2 つの新たな AMH 測定法の希釈検体を用いて精度と直線性を調べ、臨床的基準とされている BCI AMHGen II assay変法の結果と比較した。さらに、各女性の胞状卵胞数との関係についても調べた。2つの完全自動AMH assayの精度を調べたところ、1つ目の測定系の測定内の精度は0.9~1.9%、測定間の精度は2.5~ 6.5%、2つ目の測定系における測定内の精度は0.9~ 3.6%、測定間の精度は4.4~ 10.7%という結果であった。2つの測定法間の相関係数は0.991という結果であった。不妊女性群と閉経に近い女性群の新しい免疫測定法の識別能は、BCI Gen II assayと比較し有意に高い値を示し、それぞれ87.5%と68.8%であった。

Gen II(Beckman Coulter AMH Generation II assay)を用いて年齢に伴うAMHの変化を調べた。また、今回得られた値と以前のDSL assay(Diagnostics Systems Laboratory assay)で得られたノモグラムの値と比較した。臨床検査の2つの機関が調査に参加した。不妊クリニックを訪れた25~ 45歳の10,984名の新たなコホートを対象に訓練コホート5,492 名と評価コホート5,492名の 2つのコホートに分けた。また、DSL assayで測定したAMHのノモグラムの開発に参加した女性は9,601名であった。二次方程式モデルが年齢に伴うAMHの減少の状況と一致した。DSL assayで得られたノモグラムより40%の上昇するという結果は認められなかった。
