Lamellipodia is a structure that cells, those stimulatedfrom the outside, extend in the direction of motion.The cells can move forward by extending thisstructure. On the other hand, the cell peels off fromthe substrate by the contraction at the tail of the cell.In this way, the cell can move in a certain directiondue to the cycles of extension of lamellipodia in theanterior part of the cell and contraction in the tail ofthe cell.

Correlation between Rac inactivating factor FilGAP expression andprognosis in mucinous ovarian tumor

Background: FilGAP is a specific inactivating factor for Rac, which is a low molecular weight GTP bindingprotein, and binds to actin filament cross-linking protein Filamin A. In addition, FilGAP is involved in thecontrol of the cell movement mode. In recent years, the relationship between FilGAP expression and prognosisof patients with malignant lymphoma and astrocytoma were examined. Prognosis of patients with highFilGAP expression was reported to be significantly poor in the former and significantly better in the latter. Ingynecological diseases, the relationship between FilGAP expression and prognosis has not yet been fullystudied.Methods: There were 94 patients with mucous ovarian tumors (12 benign, 34 borderline and 48 malignant)who were treated at Kitasato University Hospital. We examined immunohistochemically for the expressionof FilGAP and its related factors, such as Filamin A, Integrin β1, ECT2 and a cell proliferation-related proteinKi-67. In adenocarcinoma, clinicopathological findings obtained from medical charts retrospectively andimmunohistochemical findings including FilGAP expression were correlated with the prognosis.Results: In mucinous ovarian tumors, proportion of FilGAP expression was decreased with increasing malignantpotential of the tumors, and the FilGAP expression was observed in about 80% of malignant tumors. FilGAPexpression had a significant correlation with survival outcome, and FilGAP expression, along with clinicalstage, was an independent prognostic factor for overall survival.Conclusions: Both the possibility that FilGAP's suppression effect for cellular lamellipodia formation suppressesthe extension ability and the migratory ability of cancer cells, and the possibility that the loss of FilGAPexpression ability by increasing malignant potential is related to the worsening of prognosis, were suggested.
