The mixture of betel (Piper betlel) leaf gambier (tlncaria gambi)and slaked lime (calcium hydroxide)has been used as one of the traditional medicines in Indonesia and other countries for a long time,specifically as an anti-microbial agent. Generally, the mixture of extracts is consumed by chewing in itsnatural raw condition along with many other ingredients like areca nut, cardamom and others. Thepresent studyaimedto evaluate antibacterial activityand inhibitionmechanism ofmixture ofbetel, gambierand slaked lime in a ratio of 2:r:7 on staphylococcus epidermidis with measurement on leakage of K*and c* using atomic absorption spectrometry technique, leakage of proteln and nucleic acid using lrvspectrophotometry technique and alteration of bacterial cell wall using scanning electron microscope.The concentrations of mixture used were 0.5,0.25,0.125,0.063, 0.032,0.016, 0.00g and 0.004%(w/v).The minimal inhibition concentration was about o-25%(dv) of mixture. The leakage tests of protein,nucleic acid' K* and ca2* indicated that increasing conpentration of mixture was found to enhance itsantibacterial activity. t
