INTENDED USEVit Kit® - Thaw is intended for use in assisted
reproductive procedures for warming and recovery of human oocytes
(MII) and embryos (PN to blastocyst) that have been vitrified using
Irvine Scientific’s Vitrification Freeze Kit (Catalog No.
90133-HSV, 90133-SO)
PRODUCT DESCRIPTIONThawing Solution-TS is a HEPES buffered
solution of Medium-199 containing gentamicin sulfate (35 µg/mL),
1.0 M sucrose and 20% (v/v) Dextran Serum Supplement.
Dilution Solution-DS is a HEPES buffered solution of Medium-199
containing gentamicin sulfate (35 µg/mL), 0.5M sucrose and 20%
(v/v) Dextran Serum Supplement.
Washing Solution-WS is a HEPES buffered solution of Medium-199
containing gentamicin sulfate (35 µg/mL) and 20% (v/v) Dextran
Serum Supplement *.
These three solutions are to be used in sequence according to
the step-wise microdrop warming protocol.
QUALITY ASSURANCEThe solutions in Vit Kit-Thaw are membrane
filtered and aseptically processed according to manufacturing
procedures which have been validated to meet a sterility assurance
level (SAL) of 10-3.Each lot of Vit Kit - Thaw receives the
following tests:Solutions: Endotoxin by Limulus Amebocyte Lysate
(LAL) methodology Biocompatibility by Mouse Embryo Assay (one-cell)

WARMING PROTOCOL (FOR OOCYTES AND EMBRYOS):NOTE: Procedures are
to be done at room temperature (20-27º C). DO NOT use heated
microscope stage for the following procedures. CAUTION: Minimize
exposure of specimen to light during manipulations through thawing
solutions.1. Bring the quantity to be used of TS, DS and WS to
room
temperature (20-27°C) prior to warming vitrified specimens.
NOTE: Avoid bringing the entire vials of TS,
DS and WS to room temperature repeatedly when a small quantity
of the solution is needed each time. It is better to aliquot the
quantity to be used and return the vials to 2-8°C right after
aliquoting.

USO PROGETTATOIl Vit Kit® - Thaw è previsto per l’uso nelle
procedure di riproduzione assistita per il riscaldamento e il

PROTOCOLO DE CALENTAMIENTO(PARA OOCITOS Y EMBRIONES)NOTA: todo
el protocolo debe realizarse a temperatura ambiente (20-27ºC). NO
UTILICE la platina calefactora del microscopio. PRECAUCIÓN:
Minimice la exposición de la muestra a la luz durante su
manipulación a través de las soluciones de descongelación.1. Lleve
la cantidad a utilizar de TS, DS y WS a la temperatura
ambiente (20 ºC a 27 ºC) antes de calentar los especimenes
vitrificados.
NOTA: Evite llevar repetidamente la totalidad de los viales de
las soluciones TS, DS y WS a la temperatura ambiente cuando sólo se
necesita una pequeña cantidad de solución en cada caso. Es mejor

2. Fill the liquid nitrogen reservoir with liquid nitrogen (~80
% full) and place close to the LN2 freezer containing the specimens
to be thawed.
3. Remove the canes with goblets containing the CryoTips with
vitrified specimens from liquid nitrogen storage and transfer them
into the liquid nitrogen filled reservoir.
CAUTION: Make sure that CryoTips remain submerged in LN2 (in
goblet) during transfer from storage to LN2 reservior to prevent
uncontrolled thawing of specimens.
Place the reservoir close to the microscope for rapid
manipulation.
4. Label a sterile petri dish (or lid) with necessary
information.5. Gently invert each vial of TS, DS and WS twice to
mix
contents before use.6. Prepare the dish with droplets of
solutions for warming
protocol as follows:
Aseptically dispense a sequence of 2 microdrops on an inverted
lid of sterile Petri dish as shown in Figure 1, and place the dish
on the microscope stage: • one-50 µL drop of TS • one-50 µL drop of
DS • (Two drops of WS will be set up later at step 13)
7. Place the 37°C waterbath close to the microscope. Have the
following nearby: a transfer pipet and tips, sterile sharp
scissors, Hamilton syringe and sterile wipes.
8. Using tweezers (or tongs), retrieve the specific CryoTip from
the cane in liquid nitrogen, quickly immerse the CryoTip into the
37°C waterbath (> 500 mL) and swirl gently for 3 seconds to warm
(see Figure 2) at + 24,000°C/min.
9. Quickly dispense contents of CryoTip as follows (see Figure
3):• Quickly wipe the CryoTip dry with a sterile wipe• Remove metal
cover sleeve• Cut the seal on the wide end of the CryoTip at Mark
#4• Attach wide end of the CryoTip securely to Hamilton
syringe or appropriate aspiration tool using a connector or
adaptor.
NOTE: Lift up the plunger of the syringe about 0.5 inches before
attaching the syringe to the Connector and the CryoTip.
• Gently wipe the fine tip dry with a sterile wipe. • With the
CryoTip positioned over the prepared thawing
dish, quickly cut the seal on the Mark #2 of the fine tip end
and dispense the contents of the CryoTip as a small drop (~ 1 µL)
onto a dry region of the dish above the TS drop (see Figure 4).
NOTE: AVOID BUBBLES WHILE DISPENSING THE CONTENTS.
10. Merge the TS drop with the CryoTip content and allow gradual
mixing for 1 minute (see Figure 4).
Note: the specimens will shrink and float to the top of the
drop.
NOTE: After each transfer of the specimen(s), blow out any
remaining fluid in the transfer pipet and draw up some solution
from the next drop prior to the next manipulation. Avoid creating
bubbles during the transfers.
11. Draw up some DS into the transfer pipet and transfer the
specimen(s) from the drop of TS with minimal volume to the drop of
DS for 4 minutes.
NOTE: The specimen will remain shrunken during exposure to
DS.)
DURING THIS TIME SET UP THE TWO-50 µL DROPS OF WS (WS1, WS2), AS
SHOWN IN FIGURE 4.
12. Transfer the specimen(s) to the drop of WS (WS1) for 4
minutes.
NOTE: The specimen(s) should re-expand to the original size
within 2-3 minutes in WS.
13. Then transfer the specimen(s) to the second drop of WS (WS2)
for 4 minutes.
14. Finally, transfer the specimen to a pre-equilibrated culture
dish containing the appropriate culture medium (with protein).
Incubate the specimens accordingly in a 37ºC incubator with CO2 for
3-4 hours to complete recovery prior to further manipulations (e.g.
fertilization of oocytes, transfer or extended culture of
embryos).

5. Invertire accuratamente ogni fialetta di TS, DS a WS due
volte per mescolare il contenuto prima dell’uso.
6. Preparare nel modo seguente il piatto con goccioline di
soluzione per il protocollo di riscaldamento:
Disporre asetticamente una sequenza di 2 microgocce su una tappa
invertita nel piatto di Petri sterilizzato come mostrato in Figura
1, mettere il piatto sul microscopio:• una goccia di 50 µL di TS•
una goccia di 50 µL di DS• (Due gocce di WS verranno messe dopo nel
passo 13)
7. PMettere in acqua a temperatura di 37° C vicino al
microscopio. Mantienile vicine: una pipetta di trasferimento,
forbice, Hamilton siringa e wipes sterilizzate.
8. Utilizzando le pinzette( o tongs), prendi il CryoTip
specificato dalla cana in nitrogeno liquido, immergere il CryoTip
velocemente nell’ acqua a 37° C (>500 mL) e mescolare
accuratamente per 3 secondi aspettare il caldo (vedi Figure 2) a +
24,000° C/min.
9. Dispensare velocemente i contenenti del CrypTip cosi(vedi
Figure 3):• Pulire velocemente lo CryoTip con un fazzoletto
sterilizzato.• Togliere la copertura metallica• Tagliare il
sello sulla parte grande dello CryoTip al marchio
#4.• Unire saldamente l’estremità svasata di CryoTip alla
siringa
Hamilton o ad uno strumento di aspirazione adeguato utilizzando
un connettore o un adattatore.
NOTA: Alzi il nucleo mobile della siringa circa 0.5 pollici
prima del fissaggio al connettore ed al CryoTip.
• Con il CryoTip in posizione sulla capsula pronto per il
disgelo, tagliare velocemente il sigillo e dispensare i contenenti
dello CryoTip come una goccia piccola(~1µL) su una porzione
asciutta sulla goccia di TS (vedi Figure 4).

CONTENENTI.
10. Mescolare la goccia TS con il contenuto di CryoTip e

campione dalla goccia di TS con un volume minimo al goccia di DS

esposizione al DS. DURANTE QUESTO PERIODO, PREPARARE LE DUE GOCCE

12. Trasferire il campione (s) alla prima goccia di WS (WS1) per
4 minuti.
NOTABENE: I CAMPIONI DEVONO TORNARE ALLA LARGHEZZA ORIGINALE IN
MENO DI 2-3 MINUTI IN WS.
13. Poi trasferire il campione (s) alla seconda goccia di WS
(WS2) per 4 minuti.
14. Trasferire finalmente il campione a un piatto
pre-equilibrato che contiene la sostanza di coltura appropriata
(con proteina). Incubare i campioni in accordo in una incubatrice
di 37° C con CO2 per 3 o 4 ore per completare il ricupero prima di
ulteriori manipolazioni (per esempio: la fertilizzazione di oocite,
il trasferimento o la coltura estesa degli embrioni).

7. Sitúe el baño de agua a 37ºC cerca del microscopio. Tenga
cerca el siguiente material: una pipeta de transferencia y puntas,
tijeras afiladas estériles, la Hamilton GASTIGHT® jerinuilla, y
toallitas estériles.
8. Utilizando pinzas recupere el CryoTip específico de la cesta
en nitrógeno líquido, sumérjalo rápidamente en el baño de agua a
37ºC (>500 mL) y agite suavemente durante 3 segundos para
calentamiento (ver Figura 2) a +24000ºC/min.
9. Dispense rápidamente el contenido del CryoTip de la siguiente
manera (ver Figura 3):• Seque rápidamente el CryoTip con un papel
estéril• Quite la cubierta de metal.• Corte el sello en el extremo
ancho del CryoTip en la Marca
# 4.• Fije el extremo ancho del CryoTip a la jeringa Hamilton
o
a una herramienta apropiada de aspiración mediante un conector o
adaptador.
NOTA: Levante el émbolo de la jeringuilla aproximadamente 0.5
pulgada antes de unir al conectador y al CryoTip.
• Con delicadeza, seque el CryoTip con una toallita estéril• Con
el CryoTip situado sobre la placa de descongelación
preparada, corte el sellado sobre la Marca # 2 del extremo fino
de la punta y dispense el contenido del CryoTip como un pequeña
gota sobre una zona seca de la placa por encima de la gota de TS
(ver Figura 4).
NOTA: EVITE LA FORMACIÓN DE BURBUJAS AL DISPENSAR EL
CONTENIDO.
10. Combine la gota de TS con el contenido de la CryoTip y
permita la mezcla gradual durante un (1) minuto (véase la Figura
4).
NOTA: las muestras sufrirán un pérdida de volumen y flotarán
hacia la superficie de la gota.
NOTA: después de cada transferencia de la muestra, expela el
contenido de la pipeta de transferencia y aspire un poco de la
solución de la siguiente gota antes de la siguiente manipulación.
Evite la formación de burbujas durante las transferencias.
11. Aspire un poco de DS con la pipeta de transferencia y
transfiera la muestra/s de la gota de TS con el mínimo volumen al
de la gota de DS (DS1) dejándola 4 minutos.
NOTA: la muestra permanecerá encogida durante la exposición a
DS.
DURANTE ESTE INTERVALO DISPENSE 2 GOTAS DE 50µL DE WS (WS1, WS2)
COMO SE MUESTRA EN LA FIGURA 4.
12. Transfiera la muestra/s al de la gota de WS (WS1) para 4
minutos.
(Nota: la muestra/s debe reexpanderse hasta su volumen original
en los 2-3 minutos en WS).
13. Transfiera la(s) muestra(s) a la segunda gota de WS (WS2)
por 4 minutos.
14. Finalmente, transfiera la muestra a una placa
pre-equilibrada con el medio de cultivo adecuado (con suplemento
proteico).

WARMING PROTOCOL (for oocytes and embryos):
NOTE: The warming steps include plunging the device into the
37°C TS and subsequent diluting and washing in DS and WS at room
temperature
1. Set up warming dishes (as shown in HSV device diagram):• At
37°C: Aseptically dispense 250 µl of TS into a
sterile 4-well dish or an organ culture dish and place it in a
37°C incubator without CO2 or on a heating stage at least 30
minutes prior to warming procedure
2. Identify HSV Straw(s) to be warmed from LN2 storage and
quickly transfer to LN2 filled reservoir in preparation for thawing
procedure.
3. Place LN2 reservoir close to microscope for subsequent rapid
manipulation.
4. Remove TS dish from 37°C incubator or heating stage and place
it under focus on top of the microscope stage.
5. Lift the straw enough to expose the colored handling rod.
Make sure the end with the specimen(s) remains immersed in the
LN2.
6. Hold the HSV Straw firmly between fingers, place the scissors
around the center of the colored rod and carefully score the outer
straw by pressing the scissors repetitively while rotating the
straw to perform successive incisions around the external
straw.
7. Use the scissors as tweezers to grab the capillary rod and
extract it out of the straw.
8. Immediately plunge the gutter into the 37°C TS and gently
swirl to detach specimens from device and leave it for 1
minute.
Steps 9-12 must be performed at room temperature (22-27°C).
• At room temperature: Aseptically dispense one (1) 50 µL drop
of DS on a sterile Petri dish
9. Draw up some DS into the transfer pipette and transfer the
specimen(s) from the TS dish with minimal volume to the drop of DS
for 4 minutes.
NOTE: The specimen will remain shrunken during exposure to
DS.
DURING THIS TIME SET UP THE TWO-50 µL DROPS OF WS (WS1, WS2), AS
SHOWN IN HSV DEVICE DIAGRAM.
10. Transfer the specimen(s) to the drop of WS (WS1) for 4
minutes.
NOTE: The specimen(s) should re-expand to the original size
within 2-3 minutes in WS.
11. Then transfer the specimen(s) to the second drop of WS (WS2)
for 4 minutes.
12. Finally, transfer the specimen to a pre-equilibrated culture
dish containing the appropriate culture medium (with protein).
Incubate the specimens accordingly in a 37ºC incubator with CO2 for
3-4 hours to complete recovery prior to further manipulations (e.g.
fertilization of oocytes, transfer or extended culture of
embryos).
NOTE: Recovered oocytes must be fertilized using ICSI for
optimal fertilization after
vitrification.
For additional details on the use of these products, each
laboratory should consult its own laboratory procedures and
protocols which have been specifically developed and optimized for
your individual medical program.
STORAGE INSTRUCTIONS AND STABILITYStore the unopened vials
refrigerated at 2°C to 8°C. When stored as directed, Vitrification
Freeze Kit Solutions are stable until the expiration date shown on
the vial labels.
Do not use media for more than eight (8) weeks once containers
have been opened.
As human source material is present in the product it may
develop some particulate matter during storage. This type of
particulate matter is not known to have an effect on product
performance.
PRECAUTIONS AND WARNINGSThis device is intended to be used by
staff trained in assisted reproductive procedures that include the
indicated application for which the device is intended.
Do not use any vial of solution that shows evidence of
particulate matter or cloudiness.
To avoid problems with contamination, handle using aseptic
techniques.
Vitrification Thaw Kit Solutions contain the antibiotic
gentamicin sulfate. Appropriate precautions should be taken to
ensure that the patient is not sensitized to this antibiotic.
Currently, research literature indicates the long-term effects
of vitrification on embryos remains unknown.
*Human blood derivatives used in the manufacture of this product
has been tested by FDA licensed kits, and found to be non-reactive
for the Hepatitis B surface antigen (HBsAg), antibodies to
Hepatitis C (HCV) and antibodies to Human Immunodeficiency Virus
(HIV). However, no test method offers complete assurance that
products derived from human sources are noninfectious. Handle all
Human blood derivatives as if it were capable of transmitting
infection, using universal precautions. Donors of the source
material have also been screened for risk of exposure to
Cruetzfeldt-Jakob Disease (CJD).

Minimieren Sie Kontakt (des Musters) mit Licht während Bedienung
durch Taulösungen.1. Die erforderlichen Mengen TS, DS und WS vor
Erwärmung de r v i t r i f i z ie r ten Proben au f Raumtempera tu
r bringen (20-27°C). Hinweis: Es sollte nicht der gesamte Inhalt
der TS-, DS- und WS-Ampullen wiederholt auf Raumtemperatur gebracht
werden, wenn jedes Mal nur eine kleine Menge der Lösung benötigt
wird. Es ist besser, nur die benötigte Menge abzunehmen und die
Ampullen dann unverzüglich wieder auf 2-8°C zu kühlen. 2. Füllen
Sie den Stickstoffbehälter mit flüssigem Stickstoff (~80 % voll)
und stellen Sie ihn beim LN2-Tiefkühlschrank, der die Muster
beinhaltet, die zu tauen sind.
3. Nehmen Sie die Cryocanes mit Bechern, die die CryoTips mit
verglasten Mustern beinhalten, aus dem flüssigen Stickstoff- Lager.
Versetzen Sie sie in das mit flüssigem Stickstoff gefüllte
Reservoir. VORSICHT: Sichern Sie, dass die CryoTips in LN2 (im

Bedienung.4. Etikettieren Sie eine sterile Petrischale (oder

Fläschchen von TS, DS, und WS zweimal um, um den Inhalt vor dem
Gebrauch zu mischen.6. Petrischale mit den Tropfen der Lösungen für
das Erwärmungsprotokoll wie folgt vorbereiten:Tröpfeln Sie
aseptisch eine Folge von 2 Microtröpfen auf einen umgekehrten
Deckel einer sterilen Petrischale, wie in Figur 1 gezeigt. Stellen
Sie die Schale auf den Objektträger:• Ein-50 µl Tropfen von TS•
Ein-50 µl Tropfen von DS• (Zwei Tropfen von WS werden später bei
Schritt 13 vorbereitet werden)7. Stellen Sie das 37°C Wasserbad
beim Mikroskop. Haben Sie Folgendes bereit: eine Transferpipette
und Düsen, eine sterile scharfe Schere (oder wegwerfbares
Skalpell), Hamilton spritze, und sterile Wischtücher.8. Gebrauchen
Sie eine Pinzette (oder Zange), um die spezifische CryoTip aus dem
Cryocane im flüssigen Stickstoff auszunehmen. Tauchen Sie die
CryoTip schnell ins 37°C Wasserbad (> 500 mL) ein und wirbeln
Sie das Wasserbad behutsam für 3 Sekunden, um bei + 24.000°C/Min zu
wärmen (sehen Sie Figur 2).9. Verteilen Sie schnell den Inhalt der
CryoTip wie hier angewiesen (sehen Sie Figur 3):• Wischen Sie
schnell die CryoTip mit einem sterilen Wischtuch trocken•
Metallschutzhülse entfernen• Schneiden Sie das Verschluss auf dem
weiten Ende der Cryotip (bei Zeichen #4)• Das weite Ende des
CryoTip mit einem Konnektor oder Adapter sicher an die
Hamilton-Spritze oder ein angemessenes Aspirationswerkzeug
anschließen. ANMERKUNG: Vor der Befestigung zum Stecker und zum
CryoTip heben Sie den Spulenkern der Spritze ungefähr 0.5 Zoll an.•
Wischen Sie behutsam die CryoTip mit einem sterilen Wischtuch
trocken.• Mit der CryoTip in Stellung über der vorbereiteten
Tauschale, schneiden Sie schnell den Verschluss auf Zeichen #2 des
Endes mit der feinen Düse. Verteilen Sie den Inhalt der CryoTip als

Blasen während der Versetzungen.11. Ziehen Sie ein Bisschen DS in
die Transferpipette und versetzen Sie für 4 Minute mit minimalem
Volumen das (die) Muster vom Tropfen TS zum des ersten Tropfens von
DS. (Hinweis: das Muster wird während Kontakt mit DS geschrumpft
bleiben). WÄHREND DIESER ZEIT DIE BEIDEN TROPFEN (50 μL) VON WS
(WS1, WS2) AUFSETZEN (SIEHE ABBILDUNG 4).12. Übertragen Sie die
Probe (n) auf dem ersten Tropfen von WS (WS1) für 4 Minuten.

(sollen) und als Vorbereitung für den Auftauprozess schnell in
einen LN2 –gefüllten Behälter geben. 3. Den LN2 –Behälter dicht
neben das Mikroskop stellen, damit nachfolgende Schritte schnell
ausgeführt werden können. 4. Die Schale mit der TS aus dem 37°C
-Inkubator oder von der Heizplatte nehmen und unter den Fokus auf
den Objekttisch des Mikroskops stellen.5. Den Straw so weit
anheben, dass der farbige Bedienstab sichtbar wird. Gewährleisten,
dass das Ende mit der (den) Probe(n) in LN2 eingetaucht bleibt. 6.
Den HSV-Straw fest zwischen den Fingern halten, mit der Schere um
die Mitte des farbigen Stabs greifen und die Außenseite des Straws
durch wiederholtes Andrücken der Schere unter Drehen des Straws
sorgfältig einritzen, um ein sukzessives Einritzen der
Straw-Außenseite zu gewährleisten. 7. Die Schere als Pinzette
verwenden, um das Kapillarröhrchen zu greifen und aus dem Straw
herauszuziehen. 8. Die Ablaufrinne sofort in die 37°C warme TS
tauchen und vorsichtig schwenken, um die Proben von der Vorrichtung
zu lösen und 1 Minute in der Lösung belassen.Schritte 9-12 müssen
bei Raumtemperatur durchgeführt werden (22-27°C).• Bei
Raumtemperatur: Einen (1) Tropfen DS (50 µL) mit Hilfe aseptischer
Technik auf eine sterile Petrischale bringen.9. Etwas DS in die
Übertragungspipette ziehen und die Probe(n) aus der TS-Schale mit
minimalem Volumen für 4 Minuten auf den DS-Tropfens übertragen.

medizinische Einstellung spezifisch entwickelt und optimiert worden

AUFBEWAHRUNGSANWEISUNGEN UND STABILITÄTDie ungeöffneten Ampullen
im Kühlschrank bei 2°C bis 8°C aufbewahren. Bei Aufbewahrung unter
diesen Bedingungen sind die Vitrification Freeze Kit -Lösungen bis
zum Haltbarkeitsdatum, das auf den Ampullen-Etiketten angegeben
ist, haltbar.
Die Medien nach dem Öffnen der Behälter nicht länger als acht
(8) Wochen verwenden.
Weil das Produkt menschliches Ursprungsmaterial enthält, können
während der Aufbewahrung Schwebstoffe entstehen. Diese Art
Schwebstoffe haben vermutlich keine Auswirkung auf die
Produkt-Performance.
VORSICHTSMASSNAHMEN UND WARNUNGENDiese Vorrichtung ist für die
Anwendung durch Personal vorgesehen, das in Verfahren ausgebildet
wurde, welche die für diese Vorrichtung angegebene zugelassene
Anwendung beinhalten.
Gebrauchen Sie kein Lösung-Fläschchen, das Zeichen von

Vitrification Thaw Kit Lösungen beinhalten das Antibiotikum
Gentamicinsulfat. Passende Vorsichtsmassnahmen sollen getroffen

Zurzeit zeigt Forschungsliteratur, dass die langfristigen

Blutderivate wurden mit FDA-lizenzierten Kits getestet und waren in

contenitore di LN2 (azoto liquido) e trasferirla (le) rapidamente
al serbatoio riempito dell’ LN2 (azoto liquido) in preparazione per
la procedura di scongelamento.3. Sistemare il serbatoio dell’ LN2
(azoto liquido) in vicinanza
del microscopio per poterlo manipolare susseguentemente in modo
rapido.
4. Togliere il piattino con la TS dall’incubatrice a 37 °C, o
dal piatto portaoggetti di riscaldamento, e sistemarlo sotto fuoco
sul piatto portaoggetti del microscopio.
5. Sollevare la cannuccia quanto basta per esporre l’asta di
maneggio colorata. Accertarsi che l’estremità con il campione (o i
campioni) rimanga immersa nell’ LN2 (azoto liquido).
6. Tenere la cannuccia HSV in modo fermo tra le dita, mettere le
forbici attorno alla parte centrale dell’asta colorata e marcare
con attenzione l’esterno della cannuccia premendo ripetutamente le
forbici mentre si ruota la cannuccia in modo da eseguire incisioni
successive attorno alla parte esterna della cannuccia.
7. Usare le forbici come una pinzetta per afferrare l’asta
capillare ed estrarla dalla cannuccia.
8. Immergere immediatamente l’estremità porta-campione nella TS
a 37 °C e agitare delicatamente per distaccare i campioni dal
dispositivo, quindi lasciare riposare per 1 minuto.
I punti 9-12 devono essere eseguiti a temperaturaambiente (22-27
°C).• A temperature ambiente: distribuire in modo asettico
una (1) goccia di 50 μL di DS in un piatto sterile Petri.9.
Mettere un po’ di DS nella pipetta di trasferimento e
trasferire
il campione (o i campioni) dal piattino della TS con un minimo
volume al della prima goccia di DS (DS1) per 4 minuti.
NOTA: il campione rimane contratto durante l’esposizione alla
DS.)
NEL FRATTEMPIO APPRONTARE DUE GOCCE DI 50 µL DI WS (WS1, WS2)
COME INDICATO NELLO SCHEMA DEL DISPOSITIVO HSV.
10. Trasferire i campioni nel della prima goccia di WS (WS1) per
4 minuti.
NOTABENE: I CAMPIONI DEVONO TORNARE ALLA LARGHEZZA ORIGINALE IN
MENO DI 2-3 MINUTI IN WS.
11. Poi il trasferimento goccia seconda WS campioni (WS2) per 4
minuti.
12. Trasferire finalmente il campione a un piatto
pre-equilibrato che contiene la sostanza di coltura appropriata
(con proteina). Incubare i campioni in accordo in una incubatrice
di 37° C con CO2 per 3 o 4 ore per completare il ricupero prima di

PROTOCOLO DE CALENTAMIENTO (para oocitos y embriones) NOTA: El
procedimiento de calentamiento
incluye sumergir el dispositivo dentro de la solución
descongelante TS a 37 °C y la dilución y lavado posteriores en DS y
WS a temperatura ambiente.
1. Prepare las placas de descongelación (tal como se ilustra en
el diagrama del dispositivo HSV):• A 37 °C: En condiciones de

ISTRUZIONI PER LA CONSERVAZIONE E STABILITÀConservare i flaconi
chiusi refrigerati a temperatura tra 2 °C e 8 °C. Quando sono
conservate nel modo indicato, le soluzioni del kit di congelamento
per vetrificazione sono stabili sino alla scadenza indicata sulle
loro etichette.
Una volta aperti i contenitori, non utilizzare il mezzo per un
periodo superiore a otto (8) settimane.
Nel prodotto è presente materiale di origine umana e si possono
sviluppare sostanze particellari durante la conservazione. Per
quanto noto, questo tipo di sostanze particellari non ha nessun
effetto sulle prestazioni del prodotto.
PRECAUZIONI ED AVVERTIMENTTIQuesto dispositivo è previsto per

previsto.
Non utilizzare fiale di soluzione che evidenzi qualsiasi
torbidezza o particelle di materiale
Per evitare i problemi di contaminazione, maneggiare utilizzando
tecniche in asepsi.
Le Soluzioni di Disgelo di Vetrificazione contengono
l’antibiotico solfato di gentamicina. Le precauzioni appropriate
devono essere pressi per assicurare che il paziente non è allergico
a questo antibiotico.
Attualmente, gli studi di ricerca indicano che gli effetti lungo
termine di vetrificazione sull’embrione rimangono sconosciute.
*I derivati da sangue umano usati nella fabbricazione di questo
prodotto sono stati testati tramite kit autorizzati dalla FDA e
hanno mostrato di essere non reattivi verso l’antigene di
superficie dell’epatite B (HBsAg), gli anticorpi per l’epatite C

INSTRUCCIONES DE CONSERVACIÓN Y ESTABILIDADConserve los envases
no abiertos refrigerados a una temperatura de 2 º a 8 ºC. Si se
conservan según lo indicado, las soluciones del kit de Congelación
post Vitrificación son estables hasta la fecha de caducidad
indicada en las etiquetas de los envases.
No utilice el medio durante más de ocho (8) semanas después que
los recipientes hayan sido abiertos.
Ya que el producto contiene sustancia de origen humano puede
desarrollar partículas durante el almacenaje. No hay evidencia que
partículas de esta clase tengan un efecto sobre el funcionamiento
del producto.
PRECAUCIONES Y ADVERTENCIASEste dispositivo debe ser utilizado
por personal capacitado en procedimientos que incluyan la

Las Soluciones del Kit Descongelante de Vitrificación contienen

LE MANIPOLAZIONI ATTRAVERSO LE SOLUZIONI DI DISGELO.1. Prima di
riscaldare i campioni vetrificati, portare la quantità
di TS, DS e WS da usarsi a temperatura ambiente (20-27 °C).
NOTA: evitare di portare ripetutamente a temperatura ambiente
intere fiale quando serve solo una piccola quantità di prodotto
ogni volta. È meglio prelevare solo la quantità da utilizzare e
rimettere le fiale a 2-8 °C subito dopo il prelievo.

APLICACIÓNEl kit Vit Kit® - Thaw se ha desarrollado para el
calentamiento y recuperación de oocitos humanos (MII) y embriones
(PN a blastocisto) que han sido vitrificados utilizando el kit de
congelación por vitrificación (Vit Kit™-Freeze, nº cat. 90133,
90133-HSV, 90133-SO) de Irvine Scientific en procesos de
reproducción asistida.
DESCRIPCIÓN DEL PRODUCTOSolución Descongelante-TS: es una
solución de Medio-199 tamponada con HEPES que contiene sulfato de
gentamicina (35 µg/mL), 1M de sacarosa y 20% (v/v) de Dextran Serum
Supplement*.
Solución Diluyente-DS: es una solución de Medio-199 tamponada
con HEPES que contiene sulfato de gentamicina (35 µg/mL), 0.5M de
sacarosa y 20% (v/v) de Dextran Serum Supplement*.
Solución de Lavado-WS: es una solución de Medio-199 tamponada
con HEPES que contiene sulfato de gentamicina (35 µg/mL y 20% (v/v)
de Dextran Serum Supplement*.
Estas tres soluciones deben utilizarse secuencialmente de
acuerdo a los pasos del protocolo de calentamiento en micro
gotas.
CONTROL DE CALIDADLas soluciones incluidas en el Vit Kit-Thaw
están filtradas a través de membrana y procesadas en condiciones de
esterilidad de acuerdo a los procesos de manufacturación validados

INSTRUCCIONES DE USOComponentes del Vit Kit-Thaw (por
aplicación):
• 50 µL de Solución Descongelante-TS• 50 µL de solución
diluyente-DS• 100 µL de solución de lavado-WS• 1 Conector
PROTOCOLO DE CALENTAMIENTO(PARA OOCITOS Y EMBRIONES)NOTA: todo
el protocolo debe realizarse a temperatura ambiente (20-27ºC). NO
UTILICE la platina calefactora del microscopio. PRECAUCIÓN:
Minimice la exposición de la muestra a la luz durante su
manipulación a través de las soluciones de descongelación.1. Lleve
la cantidad a utilizar de TS, DS y WS a la temperatura
ambiente (20 ºC a 27 ºC) antes de calentar los especimenes
vitrificados.
NOTA: Evite llevar repetidamente la totalidad de los viales de
las soluciones TS, DS y WS a la temperatura ambiente cuando sólo se
necesita una pequeña cantidad de solución en cada caso. Es mejor

2. Fill the liquid nitrogen reservoir with liquid nitrogen (~80
% full) and place close to the LN2 freezer containing the specimens
to be thawed.
3. Remove the canes with goblets containing the CryoTips with
vitrified specimens from liquid nitrogen storage and transfer them
into the liquid nitrogen filled reservoir.
CAUTION: Make sure that CryoTips remain submerged in LN2 (in
goblet) during transfer from storage to LN2 reservior to prevent
uncontrolled thawing of specimens.
Place the reservoir close to the microscope for rapid
manipulation.
4. Label a sterile petri dish (or lid) with necessary
information.5. Gently invert each vial of TS, DS and WS twice to
mix
contents before use.6. Prepare the dish with droplets of
solutions for warming
protocol as follows:
Aseptically dispense a sequence of 2 microdrops on an inverted
lid of sterile Petri dish as shown in Figure 1, and place the dish
on the microscope stage: • one-50 µL drop of TS • one-50 µL drop of
DS • (Two drops of WS will be set up later at step 13)
7. Place the 37°C waterbath close to the microscope. Have the
following nearby: a transfer pipet and tips, sterile sharp
scissors, Hamilton syringe and sterile wipes.
8. Using tweezers (or tongs), retrieve the specific CryoTip from
the cane in liquid nitrogen, quickly immerse the CryoTip into the
37°C waterbath (> 500 mL) and swirl gently for 3 seconds to warm
(see Figure 2) at + 24,000°C/min.
9. Quickly dispense contents of CryoTip as follows (see Figure
3):• Quickly wipe the CryoTip dry with a sterile wipe• Remove metal
cover sleeve• Cut the seal on the wide end of the CryoTip at Mark
#4• Attach wide end of the CryoTip securely to Hamilton
syringe or appropriate aspiration tool using a connector or
adaptor.
NOTE: Lift up the plunger of the syringe about 0.5 inches before
attaching the syringe to the Connector and the CryoTip.
• Gently wipe the fine tip dry with a sterile wipe. • With the
CryoTip positioned over the prepared thawing
dish, quickly cut the seal on the Mark #2 of the fine tip end
and dispense the contents of the CryoTip as a small drop (~ 1 µL)
onto a dry region of the dish above the TS drop (see Figure 4).
NOTE: AVOID BUBBLES WHILE DISPENSING THE CONTENTS.
10. Merge the TS drop with the CryoTip content and allow gradual
mixing for 1 minute (see Figure 4).
Note: the specimens will shrink and float to the top of the
drop.
NOTE: After each transfer of the specimen(s), blow out any
remaining fluid in the transfer pipet and draw up some solution
from the next drop prior to the next manipulation. Avoid creating
bubbles during the transfers.
11. Draw up some DS into the transfer pipet and transfer the
specimen(s) from the drop of TS with minimal volume to the drop of
DS for 4 minutes.
NOTE: The specimen will remain shrunken during exposure to
DS.)
DURING THIS TIME SET UP THE TWO-50 µL DROPS OF WS (WS1, WS2), AS
SHOWN IN FIGURE 4.
12. Transfer the specimen(s) to the drop of WS (WS1) for 4
minutes.
NOTE: The specimen(s) should re-expand to the original size
within 2-3 minutes in WS.
13. Then transfer the specimen(s) to the second drop of WS (WS2)
for 4 minutes.
14. Finally, transfer the specimen to a pre-equilibrated culture
dish containing the appropriate culture medium (with protein).
Incubate the specimens accordingly in a 37ºC incubator with CO2 for
3-4 hours to complete recovery prior to further manipulations (e.g.
fertilization of oocytes, transfer or extended culture of
embryos).

ATTENZIONE : Assicuratevi che Il Kit di Disgelo di Vit rimangano
sottomessi nel LN2 (goblet) durante il trasferimento dallo
stoccaggio al serbatoio LN2 per prevenire disgelo non-controllato
del campione.
Mettere il serbatoio vicino al microscopio per una rapida
manipolazione.
4. Mettere l’ etichetta sul piatto di Petri sterilizzato (o
tappo) con i dati necessari.
5. Invertire accuratamente ogni fialetta di TS, DS a WS due
volte per mescolare il contenuto prima dell’uso.
6. Preparare nel modo seguente il piatto con goccioline di
soluzione per il protocollo di riscaldamento:
Disporre asetticamente una sequenza di 2 microgocce su una tappa
invertita nel piatto di Petri sterilizzato come mostrato in Figura
1, mettere il piatto sul microscopio:• una goccia di 50 µL di TS•
una goccia di 50 µL di DS• (Due gocce di WS verranno messe dopo nel
passo 13)
7. PMettere in acqua a temperatura di 37° C vicino al
microscopio. Mantienile vicine: una pipetta di trasferimento,
forbice, Hamilton siringa e wipes sterilizzate.
8. Utilizzando le pinzette( o tongs), prendi il CryoTip
specificato dalla cana in nitrogeno liquido, immergere il CryoTip
velocemente nell’ acqua a 37° C (>500 mL) e mescolare
accuratamente per 3 secondi aspettare il caldo (vedi Figure 2) a +
24,000° C/min.
9. Dispensare velocemente i contenenti del CrypTip cosi(vedi
Figure 3):• Pulire velocemente lo CryoTip con un fazzoletto
sterilizzato.• Togliere la copertura metallica• Tagliare il
sello sulla parte grande dello CryoTip al marchio
#4.• Unire saldamente l’estremità svasata di CryoTip alla
siringa
Hamilton o ad uno strumento di aspirazione adeguato utilizzando
un connettore o un adattatore.
NOTA: Alzi il nucleo mobile della siringa circa 0.5 pollici
prima del fissaggio al connettore ed al CryoTip.
• Con il CryoTip in posizione sulla capsula pronto per il
disgelo, tagliare velocemente il sigillo e dispensare i contenenti
dello CryoTip come una goccia piccola(~1µL) su una porzione
asciutta sulla goccia di TS (vedi Figure 4).
NOTABENE: EVITARE LE BOLLE D’ARIA QUANDO SI DISPENSA I
CONTENENTI.
10. Mescolare la goccia TS con il contenuto di CryoTip e
consentire una miscelazione graduale per 1 minuto (vedere Figura
4).
NOTABENE: i campioni diminuiscono e galleggiano al superficie
della goccia.
NOTABENE: Dopo il trasferimento dei campioni, togliere qualsiasi
rimanenze di fluido nella pipetta di trasferimento e portare una
quantità di soluzione dalla goccia successive prima della prossima
manipolazione. Evitare di creare le bolle d’aria tramite i
trasferimenti.
11. Portare qualche quantità di DS nella pipetta e trasferire il
campione dalla goccia di TS con un volume minimo al goccia di DS
Per 4 minuti.
NOTABENE: Il campione rimane piccolo/diminuito durante l’
esposizione al DS. DURANTE QUESTO PERIODO, PREPARARE LE DUE GOCCE
DI 50µL DI WS(WS1, WS2), COME MOSTRATA NELLA FIGURA 4.
12. Trasferire il campione (s) alla prima goccia di WS (WS1) per
4 minuti.
NOTABENE: I CAMPIONI DEVONO TORNARE ALLA LARGHEZZA ORIGINALE IN
MENO DI 2-3 MINUTI IN WS.
13. Poi trasferire il campione (s) alla seconda goccia di WS
(WS2) per 4 minuti.
14. Trasferire finalmente il campione a un piatto
pre-equilibrato che contiene la sostanza di coltura appropriata
(con proteina). Incubare i campioni in accordo in una incubatrice
di 37° C con CO2 per 3 o 4 ore per completare il ricupero prima di
ulteriori manipolazioni (per esempio: la fertilizzazione di oocite,
il trasferimento o la coltura estesa degli embrioni).

4. Etiquete una placa de Petri estéril (o tapa) con la
información necesaria.
5. Suavemente invierta 2 veces cada vial de TS, DS y WS antes de
usarlos para mezclar su contenido.
6. Prepare la placa con gotas de soluciones para el protocolo de
calentamiento de la siguiente manera:
En condiciones de esterilidad dispense una serie de 2 microgotas
sobre la tapa invertida de una placa de Petri como se muestra en la
Figura 1, y coloque la placa en la platina del microscopio:
• 1 gota de 50 µL de TS• 1 gota de 50 µL de DS• (más tarde, en
el paso 13, se dispensarán 2 gotas de WS)
7. Sitúe el baño de agua a 37ºC cerca del microscopio. Tenga
cerca el siguiente material: una pipeta de transferencia y puntas,
tijeras afiladas estériles, la Hamilton GASTIGHT® jerinuilla, y
toallitas estériles.
8. Utilizando pinzas recupere el CryoTip específico de la cesta
en nitrógeno líquido, sumérjalo rápidamente en el baño de agua a
37ºC (>500 mL) y agite suavemente durante 3 segundos para
calentamiento (ver Figura 2) a +24000ºC/min.
9. Dispense rápidamente el contenido del CryoTip de la siguiente
manera (ver Figura 3):• Seque rápidamente el CryoTip con un papel
estéril• Quite la cubierta de metal.• Corte el sello en el extremo
ancho del CryoTip en la Marca
# 4.• Fije el extremo ancho del CryoTip a la jeringa Hamilton
o
a una herramienta apropiada de aspiración mediante un conector o
adaptador.
NOTA: Levante el émbolo de la jeringuilla aproximadamente 0.5
pulgada antes de unir al conectador y al CryoTip.
• Con delicadeza, seque el CryoTip con una toallita estéril• Con
el CryoTip situado sobre la placa de descongelación
preparada, corte el sellado sobre la Marca # 2 del extremo fino
de la punta y dispense el contenido del CryoTip como un pequeña
gota sobre una zona seca de la placa por encima de la gota de TS
(ver Figura 4).
NOTA: EVITE LA FORMACIÓN DE BURBUJAS AL DISPENSAR EL
CONTENIDO.
10. Combine la gota de TS con el contenido de la CryoTip y
permita la mezcla gradual durante un (1) minuto (véase la Figura
4).
NOTA: las muestras sufrirán un pérdida de volumen y flotarán
hacia la superficie de la gota.
NOTA: después de cada transferencia de la muestra, expela el
contenido de la pipeta de transferencia y aspire un poco de la
solución de la siguiente gota antes de la siguiente manipulación.
Evite la formación de burbujas durante las transferencias.
11. Aspire un poco de DS con la pipeta de transferencia y
transfiera la muestra/s de la gota de TS con el mínimo volumen al
de la gota de DS (DS1) dejándola 4 minutos.
NOTA: la muestra permanecerá encogida durante la exposición a
DS.
DURANTE ESTE INTERVALO DISPENSE 2 GOTAS DE 50µL DE WS (WS1, WS2)
COMO SE MUESTRA EN LA FIGURA 4.
12. Transfiera la muestra/s al de la gota de WS (WS1) para 4
minutos.
(Nota: la muestra/s debe reexpanderse hasta su volumen original
en los 2-3 minutos en WS).
13. Transfiera la(s) muestra(s) a la segunda gota de WS (WS2)
por 4 minutos.
14. Finalmente, transfiera la muestra a una placa
pre-equilibrada con el medio de cultivo adecuado (con suplemento
proteico).

WARMING PROTOCOL (for oocytes and embryos):
NOTE: The warming steps include plunging the device into the
37°C TS and subsequent diluting and washing in DS and WS at room
temperature
1. Set up warming dishes (as shown in HSV device diagram):• At
37°C: Aseptically dispense 250 µl of TS into a
sterile 4-well dish or an organ culture dish and place it in a
37°C incubator without CO2 or on a heating stage at least 30
minutes prior to warming procedure
2. Identify HSV Straw(s) to be warmed from LN2 storage and
quickly transfer to LN2 filled reservoir in preparation for thawing
procedure.
3. Place LN2 reservoir close to microscope for subsequent rapid
manipulation.
4. Remove TS dish from 37°C incubator or heating stage and place
it under focus on top of the microscope stage.
5. Lift the straw enough to expose the colored handling rod.
Make sure the end with the specimen(s) remains immersed in the
LN2.
6. Hold the HSV Straw firmly between fingers, place the scissors
around the center of the colored rod and carefully score the outer
straw by pressing the scissors repetitively while rotating the
straw to perform successive incisions around the external
straw.
7. Use the scissors as tweezers to grab the capillary rod and
extract it out of the straw.
8. Immediately plunge the gutter into the 37°C TS and gently
swirl to detach specimens from device and leave it for 1
minute.
Steps 9-12 must be performed at room temperature (22-27°C).
• At room temperature: Aseptically dispense one (1) 50 µL drop
of DS on a sterile Petri dish
9. Draw up some DS into the transfer pipette and transfer the
specimen(s) from the TS dish with minimal volume to the drop of DS
for 4 minutes.
NOTE: The specimen will remain shrunken during exposure to
DS.
DURING THIS TIME SET UP THE TWO-50 µL DROPS OF WS (WS1, WS2), AS
SHOWN IN HSV DEVICE DIAGRAM.
10. Transfer the specimen(s) to the drop of WS (WS1) for 4
minutes.
NOTE: The specimen(s) should re-expand to the original size
within 2-3 minutes in WS.
11. Then transfer the specimen(s) to the second drop of WS (WS2)
for 4 minutes.
12. Finally, transfer the specimen to a pre-equilibrated culture
dish containing the appropriate culture medium (with protein).
