A study included testing 20 subjects using a cream

containing 2% apple stem cell extract twice daily for

4 weeks. All subjects showed a significant reduction in

wrinkle depth of at least 15%.

Cells Human keratinocyte progenitor cells Assay A novel Progenitor Cell Targeting technology was used to isolate human epidermal stem cells. The method consists essentially of culturing primary human keratinocytes in a medium specifically designed to mimic the environment of the in vivo stem cell niche. This special, fully defined cell culture medium leads to an enrichment of so called keratinocyte progenitor cells (stem cells). Compared to freshly isolated cells, the cell population of passage 4 is characterized by a 10-fold increase of

CD34/alpha6 Integrin double labelled cells. CD34 and alpha6 Integrin are known markers of epidermal stem cells. These keratinocyte progenitor cells were incubated with the Vitis vinifera stem cell extract and were UV-light exposed for 10 minutes at 450 kJ/m2 using the Suntester CPS+. Not radiated and not treated cells served as a control. The colony forming efficiency (CFE) was determined. For analysis of CFE, cells are seeded at low density. The number of colonies formed is a value for the concentration of active progenitor/stem cells. Test parameters UV light exposition: at 450 kJ/m2 using the Suntester CPS+ Epidermal stem cell markers: CD34 and alpha6 Integrin Results Vitis vinifera stem cell extract was found to protect human keratinocyte progenitor cells against UV radiation and strongly stimulate the CFE irrespective of the cells being radiated or not.
