Throughput for transporters The SURFE2R 96SE is designed for activity measurement of electrogenic membrane transporters and pumps, which generate currents too low to resolve by patch clamp or which are not expressed in membranes accessible for patch clamp (e.g. bacterial, organelles). The technology enables label- and radioactivity-free real time measurements with high time resolution. The SURFE2R 96SE is a fully parallel 96 well based system, generating 96 data points in about 5 minutes. The highly specialized software tools SURFControl and DataControl and a set of optimized workflow modules enable the automatization of the experiment starting with the preparation of the measuring sensor to export and data analysis.

High flexibility The SURFE2R 96SE employs Solid Supported Membrane (SSM)-based electrophysiology, which enables characterization of substrates, inhibitors, modulators and cofactors. Possible targets range from electrogenic membrane transporters like symporters, exchangers or uniporters to ion pumps and ligand-gated ion channels. Membrane fragments derived from cells, organelles, and tissues or liposomes containing the purified target protein can serve as test sample. The SURFE2R 96SE provides a very flexible assay design. Single dose experiments, cumulative IC50 or EC50 determinations or more complex workflows with duplicates and control sequences can be designed by the user.

Automatic data analysis SURFE2R 96SE experiment results are analyzed with DataControl, a Nanion developed software allowing straightforward and fast evaluation of large data volumes – the basis for high throughput experiments. DataControl provides flexible tools for compound handling, peak and area determination, calculations and curve fitting. The use of templates enables fully automated processing of experimental data. Export of analyzed or raw data to other software tools, e.g. Genedata Screener®, allows individual data handling and processing.

The automated sensor preparation can be performed independently from the measurements by using other automated liquid handling platforms.

Each measurement takes about 5 min. One typical measurement sequence contains three measure-ments: (1) negative control, (2) full activation and (3) inhibition. 3 x 96 datasets therefore take 15 min.

In this scenario 25 plates can be measured per day (8 h) yielding 25 x 3 x 96 = 7200 datasets.

“The SURFE2R makes extremely challenging electrophysiological targets accessible to robust routine analyses. For example, we have developed an assay for investigating nicotinic acetylcholine receptor a7 ion channels, using membranes from the Pacific electric ray eel, Torpedo California. In this way, we overcome many of the well-known difficulties associa-ted with this ion channel, which allows us to efficiently obtain information regarding com-pound pharmacology. Obtained pharmacology values match those of patch clamp re-cordings exceptionally well.
