Summary

Multiple sirtuins, including SIRT1 and SIRT3, are likely to play roles in critical injuries

A major role of sirtuins is to suppress the inflammatory response in macrophages

Timing and cell-type specificity of interventions will likely be critical…genetic models are extremely useful to tease these details out

Translation

Resveratrol and NMN could be moved into larger animal models after a few additional studies to address timing and optimal dosing.

There is potential to greatly improve the efficacy of these interventions by using genetic tools to understand the mechanisms.
