Section1Introduction
ThismanualisintendedtoassisttheuserwiththebasicfeaturesoftheQueenStationtminstrumentalinseminationdevice.Thismanual,however,isnotintendedtobeatrainingmanualon“how”toinseminatequeens.Itisincumbentupontheusertoattendatrainingsessiononqueeninsemination.
Thisinstrumentrepresentsthousandsofhoursofresearchanddevelopmentincludinginputfromuniversity,professionalandqueenbreedingorganizations.Youcanbeassuredthatyouhavepurchasedthefinestinstrumentavailableanywhereintheworldtoday.
TheQueenStationtmisvirtuallymaintenancefreeandwithpropercleaning,useandstorageitwilllastalifetime.Someofthefeaturesincludedwiththisdeviceareuniqueandareunderpatentpendingprotection.
TheadvantagesoftheQueenStationtmarelistedhere:
1. LargeindustrialCo2cylindersarenolongerneeded.Ifyoutraveltoinseminatequeensyoucantakethe74gCo2cartridgesandnothavetoworryabouttheavailabilityofCo2.TheindustrialCo2cylindersareinherentlydangerousandhaveseriouslyinjuredpeople.Wehavesuccessfullyinseminatedmorethan100queensonone74gramcartridgeandhadenoughCo2remaininginthecartridgeforanother100queens.
2. Theon-boardgoosenecklightsarestate-of-the-artunitsthatarecontrolledwithamicroprocessordimmersystem.Theselightscompletelyreplacethebulkyandexpensiveunitscurrentlyneededwithdevicesofferedbyothervendors.Theselightsruncoolandcanbepositionedwherevertheuserdesires.
3. ABubblecounterthatactuallyallowstheusertocountaccuratelysizedbubbles.Withthisinstrumentthereisnolongeranyneedforglassbeakers,hoses,stoppersandclutterassociatedwitholdersystems.OldersystemsusingbeakerscannotcontrolthebubblesizeandhencecannotcontrolthetrueamountofCo2beingdispensedtothequeenholder.
4. Superiorsyringestage(slide).Allothercommerciallyavailableunitsfeatureaninferiorsyringestage.ThesyringestageontheQueenStationtmisalaboratorygradeunitthatallowstheusertoadjustthetensionasheorsheseefit.
5. Ballsocketsthatpresenteventensioninallareasofrotation.Otherunitshaveaseriousflawthatcausesthetensiononthedorsalandventralhookstobehigherinonedirectionofrotationthantheother.Ourunithassmooth,eventensioninalldirections

makingthedamageofqueensunlikely.
6. Fixeddorsalandventralhooklocationsrelativetothequeenholder.Otherunitshaveadjustablepoststhatholdthehooks.Thiscausesissueswithoperatorswhomaynotbeabletofindtheperfectlocationandangleneededtoinseminatethequeen.Withfixed
7. Boomtrinocularmicroscopebuiltintotheunit.Thissavesspaceontheworkbenchandeliminatesyetanothersetupproblemfortheoperator.
8. Hardanodizedfinishonmostparts.Hardanodizeisharderthanstainlesssteel,dielectricandstainresistant.Thisequatestoeaseofcleaningandmaintenance.
9. Optionalhardshippingandstoragecontainermakestransportingtheuniteasy.

AftercarefullyunpackingallcomponentsfromPelicancase,placethebaseunitinthelocationwhereyouwillbeinseminatingqueens.Note:thebaseunithasthreeSorbothanevibrationisolationpadsonthebottom.Thesepads,whennew,areverystickyandassuchmakeitdifficulttoshifttheunit.Placingitinitsproperlocationisthebestpractice.Onceinproperlocation,placethemicroscopeboomcolumn(threadsfirst)intotheshaftlocklocatedattherearofthebase.Presstheshaftlockclosed

Section3CareandCleaning
1. Base:Cleaningthebaseisextremelyeasy.Afterdisassemblyforstoragetheusershoulduseeitherethanol,isopropylalcoholoranalcoholbasedwindowcleanersuchasWindex.Useacleancottonclothandspraythealcoholonthecloth(notonthebase)andwipetheentireunit.
2. Microscope:Themicroscopeshouldbecleanedinthesamemannerasthebase.Themicroscopelensesandeyepiecesshouldbewipedcleanbeforestoring.
3. BubbleCounter:Thebubblecounterexteriorcanbecleanedwithethanoloralcoholonacleanragbutbesuretowipethepolycarbonatetubewithadrycloth.Thiswillpreventthealcoholfromcrazingthesurfaceofthetube.
4. SyringeStage(slide):Theexteriorofthestagecanbecleanedwiththeethanoloralcoholclothbutitshouldbenotedthatitispossibletoalsocleantheoilfromthedovetails.Aftercleaningwithalcoholcloththeusershouldre-lubricatethedovetailsasfollows.Rotatetheslidecompletelytooneendofitsrange.Addonedropoflubricatingoil(suchas3-in-Oneoil)oneachdovetail.Slowlyrotatetheslidetotheotherendofitsrange.Wipeanyexcessoilfromthestage.Returntheslidetoitscenteroftravel

5. DorsalandVentralHooks:Priortoeveryuse,aftereveryuseandin-betweeneveryqueeninseminationthedorsalandventralhooksshouldbesterilized.Sterilizationcanbeaccomplishedsterilealcoholwipes.Experiencedqueeninseminatorswilltellyouthatoneofthemostlikelysourcesoffailedinseminationisaresultofinfectionfromcontaminatedhooksandsyringes.
6. Syringe:Dependingonwhichtypeofsyringetheuserpreferswilldeterminethepropercleaningmethod.ManyuserspreferthelargerHarbostylesyringewhileotherspreferthesmallersyringethatinstallsdirectlyintothesyringeholderontheQueenStationtm.
Thesyringeshouldbecompletelydisassembledandwipedwithasterilealcoholwipe.Thesyringeshouldbereassembledandtheexteriorwipedclean.Thesyringeprovidedwithyourinstrumentisa1mldisposableunitandshouldnotbecleaned.
7. Lights:Theexteriorofthelightscanbewipedwiththealcoholcloth.
Section4Co2System
TheintegratedCo2systemisoneofthesignificantfeaturesoftheQueenStationtmandneedstobefullyunderstoodandmanaged.FailuretounderstandtheCo2systemwillbeasourceofproblemsfortheuser.Ifyouastheuserisusedtousinga100cubicfootindustrialcylinderyoumayhavedevelopedahabittonotbeconcernedabouttheamountofCo2availabletoyou.YouwillquicklybecomecognizantofCo2conservationwiththissystem.Itiscriticaltofollowthestepslistedheretobesuccessful.
1. CartridgeInstallation/Removal:Priortoinstallingorremovingthecartridgeshutoffallthreecontrolvalves.Useonly74gCo2cartridge.ItisrecommendedtouseONLY74gramcartridgesthatareintendedforbeveragedispensingsystems.DONOTUSECo2CARTRIDGESINTENDEDFORPAINTBALLGUNSORFISHTANKS.74gCo2cartridgesintendedforbeverageservicecontainfoodgradeCo2andwilldoyourqueensnolastingharm.AsidefromthequalityoftheCo2containedinthecartridge,thethreadsandsealsintheQueenStationtmregulatormaynotmatchothercartridges.Sparecylinderscanbeobtainedlocallyfrombrewerystores,on-linefrombrewerystoresordirectlyfromApisEngineering.

InstallingtheCo2cartridgeisstraightforward.Simplyscrewthecartridgeintothethreadsontheregulatorbody.ThereisasmallprojectioninsidetheCo2regulatorbodythatpiercesthesealoverthecartridge.Itisimportantthatthecartridgebescrewedoncompletelyandhandtightwithoutstoppingthethreadingmotion.Iftheuserinterruptsthethreadingmotionitispossibletocausethegastoescapebeforethecartridgeisfullyseatedagainsttheseal.Oncethecartridgeisinstalleditshouldnotberemoveduntilitisempty.IfthereisCo2gasremaininginthecartridgeanditisremovedthegaswillescapeandthecartridgewillcompletelyempty.Itcannotbesealedandreused.Thecylinder,ifremovedwithCo2gasremaining,willnotpresentadangerexceptthaticemayformontheexteriorofthecartridge.Ifyoupurchasedtheoptionaltravelcasethefoamlinerhasbeendesignedtoprotectthecylinderwithitremainingontheregulatorbody.
2. WaterFill:Withallthreecontrolvalvesclosedthebubblecountercanbefilledwithwater.Note:onlydistilledwatershouldbeusedinthebubblecounter.Distilledandde-ionizedwaterisacceptablebutnotrequired.Thebubblecounterisfilledbyremovingthesmallthumbscrewonthesideofthepolycarbonatetube.Includedwithyourinstrumentisa3mlblunttippedsyringe.Fillthissyringewithdistilledwaterandthenfillthebubblecounterthroughthethumbscrewhole.Fillthepolycarbonatetubetoapproximately1/8”belowthethumbscrewhole.ReplacethescrewmakingsurethesmallO-ringisstillonthescrew.Fromtime-to-timethebubblecounterwillneedrefilled.ThisisbecauseminuteamountsofthewaterbindtotheCo2andiscarriedoutofthesystem.Whentheuserneedsto“topoff”thesystemallvalvesneedtobeclosedandthesystemrechargedwithCo2asdescribedbelow.
3. PressureAdjustmentProcedure:InordertoeliminatewastingCo2andhenceprematuredepletionofCo2cartridgethefollowingproceduremustbeadheredto.
a. Checktomakesureallthreecontrolvalvesareclosed.Thevalveswillbeoffifthestep#1(above)wasfollowed.
b. AfterinstallationofCo2cartridgeturntheregulatorknobclockwisetoanindicatedpressureof5psiasshownonthedial.Atnotimeismorethan5psirequiredtoanesthetizequeens.Afteranyofthethreevalveshavebeenopenedtheuserneedstowait15secondsbeforeopeningthenextvalve.This15seconddelayallowsthepressuretostabilizewithinthesystem.Keepinmindyourgoalistoproducea3bubblepersecond(bps)flowrate.ThisisanextremelysmallvolumeofCo2gasandittakestimetofillthelines.ItshouldbenotedthatCo2isheavierthanairandthisworkstoyouradvantagewithinthe

systembecauseallairwillbedrivenoutofthelines.Bepatienthere.
c. Afterwaiting15secondsthenextvalvetoopenistheflowregulator.Thisisthebigblackknobwiththeredlockring.Theredlockringneedstobeliftedupinordertoadjusttheknob.Raisetheredringandopentheknob(turncounterclockwise)onefullturn.Pushtheredlockringbackdown.Wait15secondsagain.OpeningthisvalveallowstheCo2gastoflowthruthelinestothebubblecounter.Atthispointthebubblecounterispressurizedbutnobubblesareobservablebecausetheflowcontrolvalveontopofthebubblecounterisclosed.Bubblescanonlyappearoncethisvalveisopened.
d. Thissectionisthemostcriticaladjustment.Slowlyopenthevalveontopofthebubblecounterbyturningcounterclockwise.Ifthisvalveisopenedtoquicklythebuiltuppressureinthelineswilldriveallofthewateroutofthebubblecounterandintothelinesfeedingthequeenholder.AfterasmallopeningofthevalveWAITtoseeifanybubblesappear.Oncebubblesappearthevalveonthetopofthebubblecountercanbeadjustedforapproximately3bps.Atthispointyoumayrecallreadinginotherpublicationsthat8-10bpsisappropriateandinothersyoumayhavereadthatcontinuousCo2isrequired.Whythedifferences?Itsimplyamatterofbubblesize.Thebubblesgeneratedinthissystemflowthroughapreciseorificethatcontrolsbubblesizeand3bpsisadequate.
e. Ifafteropeningthevalveonthetopofthebubblecounterdoesn’tproduceanybubblesthenclosethisvalvecompletelyandgobacktotheflowcontrolvalveandliftuptheredringandopenthisvalveanother½turn.Pushtheredlockringdownandrepeatsection(d).
4. DrainingBubbleCounter:Occasionallyitmaybenecessarytodrainthesystem.a. ShippingorLongTermStorageofInstrument:Iftheuserneedstoshipthe
instrumentviaanymethodi.e.airtransportorgroundthebubblecountershouldbedrained.Additionally,iftheinstrumentistobestoredforlongperiodsitshouldbedrained.Thisisneededfortworeasons.1)Ifyoushipyourinstrumentitwilllikelybeturnedonitssideorupsidedownandthewaterwillleakout.And2)Longtermstorageandshipmentmayexposetheinstrumenttofreezingconditions.
b. DrainingtheBubbleCounter:Iftheinstrumentneedsdraineditishighlylikelythatyoualsoneedtodisassembleittoo.Inanyevent,todrainthesystemtheinstrumentneedstobedisassembled.Oncedisassembledremovethesmallthumbscrewinthesideofthebubblecounterandturntheinstrumentupsidedownandallowthewatertodrainoutofthethumbscrewhole.

5. TemporarilyShuttingOffCo2Flow
a. BetweenQueens:AftercompletingtheinseminationofaqueeniftheuserisgoingtoimmediatelyinseminateanotherqueennothingneedstobedonetoshutofftheCo2flow.If,however,thereisgoingtobesomelengthoftime(morethan3minutes)itisanadvantagetoshutdowntheCo2flow.TotemporarilystopCo2flowsimplyclosetheflowcontrolvalveontopofthebubblecounter.Asyouturnthevalvetrytorememberhowmanyturnsitrequiredtocompletelyclosethevalve.Thiswillmakeiteasiertorestoreflowifyouhaveanideaofwherethevalvewaspreviouslyset.Note:WhenyouneedtoopenthevalvetorestoreCo2flow,openthevalveslowlyandinsmallincrementswithapausebetweeneachadjustment.Insummary,forshorttermCo2flowstoppagesallofthevalvesdonotneedtobeclosedonlythevalveontopofthebubblecounter.
6. LongTermShuttingOffCo2Flowa. FinishedUsingtheInstrument:Uponcompletionofinseminationofqueensit
willbenecessarytoshutoffCo2flow.Thisisaccomplishedbyclosingallthreevalves.ClosethevalveontheCo2regulatorfirst.ThisisaccomplishedbyturningtheknobCounterclockwise.Oncethisvalveisclosedthepressuregaugewilldroptozero.Next,lifttheredringontheflowcontrolvalveandclosethevalvebyturningtheknobClockwise.Finally,closethevalveontopofthebubblecounterbyturningtheknobClockwise.
Section5InstallingQueeninQueenHolder:Includedwithyouinstrumentaretwoplastictubes.Oneofthetubesistaperedononeendandtheotherissquareonbothends.Thetaperedtubeisusedtoholdthequeenduringanesthetizationandinseminationanditisalsousedtotrapthequeenpriortoinsemination.Thetubewithoutthetaperedend,sometimesreferredtoasthe“back-up”tube,isusedtocausethequeentobackupintothetaperedtube.Onceyou’veselectedaqueenforinseminationplaceherinthesquarecuttube.Oncesheisinthetubeplacethesquareendofthetaperedtubeagainsttheothertubeattheendofthetubethathasthequeens’stingshowing.Bepatientandshewillbackupintothetaperedtube.Placethetubeonthequeenholderpostontheinstrument,whichshouldhaveCo2flowing,andshewillbeanesthetizedandreadyforinsemination.

Section6SecondaryAnesthetizationofQueens
Empiricalevidencehasshownthatsecondaryanesthetizationofqueenscausesthemtobeginlayingsoonerandforlongerperiods.Yourinstruments’accessorykitboxdoublesasthesecondarysystem.TheboxhasventholesinthelidandaCo2portonitsside.AlsoincludedisaconnectorhosethatistobeattachedtothequeenholderpostontheQueenStationtmandthesecondarybox.Thesecondaryanesthetizationboxwillholdsevenoftheso-calledCaliforniaMinicages.Theflowrateforthesecondaryboxismaintainedat3bps.Thisratewillfilltheboxandforcetheairoutthroughtheholesinthelid.

Section7LightingSystem
TheQueenStationtmincludesabuiltinlightingsystemthatiscontrolledbyamicroprocessordimmersystem.Thissystemcompletelyeliminatesthecostlyandspacewastingindependentsystems.
1. Installation/Removal:Thelightsareeasilyinstalledbypressingthe3pinbaseintothesocket.Itdoesnotmatterwhichlightgoesinwhichsocket.Toremovethelightspressthetabonthefrontedgeofthebasemountedconnectorandpulluponthebaseofthelamp.Note,donotpullupontheflexibleshaft.
2. Alignment:Thealignmentofthelightsisentirelydecidedbytheuserasregardswheretheuseriscomfortablewiththeillumination.Therecommendedmethodisasshowninthephotos(figure5and6).Bycurlingthelightsaroundthemicroscopeknobsasshowitkeepsthelightsoutofthewaywhenadjustingthemicroscopeandthesyringeslide.
3. Adjustment:Thelights’intensityisadjustedusingthepotentiometerontherightsideofthebase.Turningtheknobcompletelycounterclockwiseturnsthelightsoff.Turningtheknobclockwiseincreasestheintensity.Notethatbothlightsmustbeinstalledonthebaseforproperoperation.
