Executive Summary African Penguin (Spheniscus demersus)
The African Penguin SSP population at the time of analyses
consists of 935 (503.429.3) individuals at 50 AZA institutions. A
target size of 1100 individuals was designated for this population
by the Penguin Taxon Advisory Group (TAG) in their 2015 – 2020
Regional Collection Plan (RCP). Under AZA’s current sustainability
designations, this population qualifies as a Green SSP.
The genetic statistics reported in this plan are based on an
analytical studbook and reflect the exclusion of 167 recently
identified potential hybrids that would have otherwise been
included as potential breeders. The currently living 714 birds
remaining in the potentially breeding population are descended from
94 founders with no additional potential founders remaining.
Despite the exclusion of many extra birds in 2016, gene diversity
in the population is still very high (98.62%) and equivalent to
that found in about 36 unrelated individuals (FGE = 36.18).
Furthermore, the temporary loss of these birds as potential
breeders has not significantly impacted long term genetic
projections, which indicate population gene diversity is at 97% in
100 years and above 95% for over 200 years. Gene diversity of 90%
is a standard benchmark for AZA managed populations, and below this
level of gene diversity it is expected that reproduction will be
increasingly compromised by, among other factors, lower hatch
weights, smaller clutch sizes, and greater hatchling mortality.

In order to reach the TAG recommended target size of 1100 in
three years (λ=1.053), approximately 126 to 152 hatches are needed.
Only approximately 68 hatches are needed to maintain current
population size (λ=1.00). With an average of 76.4 hatches recorded
in the SSP over the past five years, this population is expected to
grow but at a slower rate than is needed to fill SSP spaces by
2020. With an additional 77 viable females excluded from breeding
this year due to potential hybridization, SSP institutions will
need to increase breeding efforts with the remaining recommended
females in order to meet demographic needs.
As with most SSP populations, recommended pairings are
prioritized to maintain or increase gene diversity through
considerations of mean kinship, avoidance of inbreeding,
differences in sire and dam mean kinships, and the degree of
uncertainty within a pedigree. Every effort was made to keep
genetically valuable pairs together and at their current
institutions. Additionally, the SSP considered the hypothetical
genetic value of potential hybrids and avoided breaking up pairs or
moving birds based on their potential hybrid status alone.
Summary Actions: The SSP recommends 174 breeding females, with
79 of these encouraged to double-clutch. MateRx
matrices have been provided for several large colonies to help
managers in identifying valuable pairs and regulating reproduction.
Approximately 65 transfers have been recommended in order to
establish new colonies, create new pairs, and meet institutional
needs. Any and all future transactions not listed in these current
recommendations must be pre-approved by the SSP Steering Committee,
including the transfer of offspring produced as a recommendation of
the current plan.

Description of Population Status
African Penguin (Spheniscus demersus) SSP
Introduction: The African Penguin SSP population at the time of
analyses consists of 935 (503.429.3)
individuals at 50 AZA institutions. A target size of 1100
individuals was designated for this population by the
Penguin Taxon Advisory Group (TAG) in their 2015 – 2020 Regional
Collection Plan (RCP). Under AZA’s
current sustainability designations, this population qualifies
as a Green SSP.
The African Penguin Species Survival Plan (SSP) was initiated in
1996, and breeding and transfer
recommendations have been produced every two years since 1997.
However, the recommendations
presented in this current plan will be reevaluated in a single
year’s time following molecular investigation into
potential hybridization in the SSP. Comprehensive genetic and
demographic analyses of the North American
Regional African Penguin Studbook (current to 1 April 2016) were
performed in July 2016 using PopLink 2.4
and PMx Version 1.3.20160601. Recommendations contained in this
Breeding and Transfer plan supersede
those made by earlier plans.
Status and Conservation: IUCN (2012): Endangered; CITES:
Appendix II; USFWS: Endangered Analytical Population: This
population currently has an 87.9% known ancestral pedigree. An
analytical
overlay was created to address unknowns in the pedigree, based
on initial assumptions previously developed
by Bob Lacy and additional assumptions created at previous SSP
meetings (Appendix A). Fifty-four (20.34.0)
birds were excluded from the genetic analyses due to reasons
including age, participation in education
programs, medical concerns, behavioral issues, and unresolved
pedigree (Appendix C). Additionally, 167
(90.77.0) individuals that would have otherwise been included
are excluded as potential breeders this year
until molecular analysis can confirm species purity. For more
information on management of potential hybrids
in the SSP, please see page 7. Following these pedigree
assumptions and exclusions, the potentially
breeding population consists of 714 (393. 318.3) individuals
with a 100% known pedigree.
Demography: The African Penguin SSP population exhibited rapid
growth from the early 1970s to the late
1980s due to successful breeding by zoos and aquariums (Figure
1). Growth rates during this time ranged
from lambda (λ) of 0.90 to 1.82. Starting in the 1990s and
coinciding with SSP management, annual growth
rates due to zoo and aquarium hatches were more modest but with
a general trend of positive growth (mean
λ over past 26 years = 1.033). In the last five years, the
population has grown on average by about 3% (λ =
1.034). In order to reach the TAG recommended target size of
1100 in three years (λ=1.053), approximately
126 to 152 hatches are needed. Only approximately 68 hatches are
needed to maintain current population
size (λ=1.00). With an average of 76.4 hatches recorded in the
SSP over the past five years, this population is

Age distribution
The age structure of the total African Penguin SSP population
appears fairly stable, with no empty age
classes and a large base of juveniles to support future
reproduction and demographic stability (Fig. 2, left).
Even after the exclusion of 221 birds, 23% of the total
population, the potentially breeding population still
approximates a stable distribution with many breeding males and
females (Fig. 2, right). In both cases, a male
sex bias is evident, which could potentially disrupt breeding in
this monogamously breeding species;
however, unpaired males can be managed in single sexed colonies
when necessary. Due to the large size of
this population and the specie’s long reproductive lifespan,
this is not a major concern currently but should
continue to be monitored in the future.
Longevity
According to studbook data in AZA between 1970 to present, if an
African penguin in AZA survives past the
first year of life, median life expectancy is 18.6 years for
males and 15.3 years for females (Appendix F). This
indicates that about half of animals hatched in AZA that survive
past the first year of life are expected to die
before these ages and about half after. Although the studbook
also contains records of African Penguins
living upwards of 41 years old for both sexes, median life
expectancy is a more appropriate way to
communicate the typical lifespan of this species in AZA zoos.
The studbook also contains a record for a wild
hatched male living to 45 years (with a +/- year estimate);
however, because hatch dates and ages at entry
into zoos are unknown and difficult to verify, 41 years should
be viewed as a more reliable maximum
longevity until more data accumulate. First year mortality
observed for this species is approximately 32% for
males and 31% for females (Appendix D). As with most egg laying
species, only eggs which hatch and are
recorded in the studbook are reflected in these mortality
rates.
Reproduction
Reproduction has been recorded as early as 1.5 years for both
sexes, though first reproduction at around two
years is more typical. Peak female reproductive range, based on
studbook data in AZA from 1970 to present,
occurs between four and 15 years, though a decline in fecundity
is noticed after eight years. Nonetheless,
females have reproduced at as old as 32 years. Males have a
longer reproductive span, with the oldest sire
recorded at 35 years and fairly consistent fecundity rates
observed between four and 28 years. However,
decline in fecundity is evident after age class 14. Reproduction
has been recorded year round. Clutch size
ranges from one to two (mean = 1.4) following an approximately
38 day incubation period. Multiple clutches in
a single season are possible and will be needed to meet
demographic needs this year.

Genetics: These genetic statistics are based on an analytical
studbook with pedigree assumptions intended
to estimate relatedness. They also reflect the exclusion in 2016
of 167 recently identified potential hybrids that
would have otherwise been included as potential breeders. These
animals will remain excluded until species
purity can be verified through molecular genetic analysis.
Consequently, population statistics reported in this
plan may not be comparable to previous years and could change
again in the future.
The currently living 714 birds remaining in the
potentially breeding population are descended
from 94 founders with no additional potential
founders remaining. Despite the exclusion of
many extra birds in 2016, gene diversity in the
population is still very high (98.62%) and
equivalent to that found in about 36 unrelated
individuals (FGE = 36.18). Furthermore, the

breeding animals with low and well matched mean kinships and
through maintenance of the already high

this time, the importation of new founders is not
necessary to meet genetic goals.

Management of hybridization: Molecular genetic testing recently
confirmed an African x Humboldt hybrid
penguin in the European Endangered Species Programme (EEP). The
original hybridization event is
suspected to have occurred before 1982 in STUTTGART, which held
African and Humboldt penguins in the
same enclosure until 1981. Consequently, any individual with
STUTTGART ancestry is considered to have a
high hybridization risk. In order to identify other potential
hybridization events, the EEP is investigating historic
mixed colonies of African and Humboldt Penguins through
comparative analysis of their respective EEP
studbooks (Bos, Schad, & Fienieg, 2016).
Through this comparative analysis, the EEP identified 20
individuals exported to the SSP that have ancestry tracing
back
to mixed exhibits in the EEP. The SSP imported these
individuals during two events, the first in 1984 from
AMSTERDAM to BALTIMORE and the second in 2006 from
LISBON to GA AQUARIUM. Of these 20 imports, 14 have
currently living descendants in the SSP. Ten of the living
descendants are permanent non-breeders, which means that
167 birds that would otherwise have been included as
potential
breeders are excluded this year. These constitutes
approximately 18% of the total SSP population (167/935; Fig.
4).
However, due to its large size, the SSP population can afford
to
exclude reproductively viable potential hybrids as breeders in
the
short term until molecular analyses can confirm species purity.
It
should be noted that the fact that these birds trace back to
mixed
African and Humboldt penguin exhibits in Europe only indicates
potential of hybridization not confirmation.
The SSP is currently working to identify the most feasible and
cost effective way to collect blood samples for
analysis. Any potential hybrid confirmed to be purebred will be
added back into the potentially breeding
population in the future. In these current recommendations, the
SSP considered the hypothetical genetic
value of potential hybrids and avoided breaking up pairs or
moving birds based on their potential hybrid status
alone.

In order to meet demographic goals, some demographic pairs,
consisting of individuals with higher than
average mean kinships and/or low levels of relatedness, have
been recommended. The SSP made every
attempt to avoid breeding between genetically mismatched
individuals, which would only flood the population
with common alleles, permanently link rare and common lineages,
and make pairing recommendations
increasingly difficult in the future.
The studbook keeper will request an update from all breeding
facilities in approximately one year, and the
management group will closely monitor the breeding success and
make adjustments if necessary. Revised
recommendations may be distributed in 2017 if the SSP is able to
resolve questionable species purity by that
time.

1. The SSP recommends 174 breeding females among 37
institutions.
a. These breeding recommendations apply to the upcoming year and
may be revised again by
the end of 2017 if questions concerning the potential hybrid
issue have been resolved by this
time. For more information on how the SSP is managing potential
hybrids in the population,
please refer to page 8.
b. Approximately 79 recommended breeding pairs may double clutch
in the upcoming year.
c. Six institutions with large colonies have been provided with
MateRx matrices in order to help
them to work with the SSP Coordinator in identifying genetically
valuable pairs and regulating
reproduction. For more information on using MateRx, see page 10.
Any breeding pairs with
MSI scores of 1, 2, or 3 are recommended to double clutch.
However, contact the SSP
Coordinators first for approval to breed any potential pairs
using the MateRx matrix.
2. The SSP recommends 65 transfers in order to create new
breeding pairs and to meet institutional
needs. An additional five animals are available currently for
placement.
3. Attempts were made to address existing pairs with specific
recommendations. The SSP considered
the hypothetical genetic value of potential hybrids and avoided
breaking up pairs or moving birds based on their potential hybrid
status alone. Institutions should consult with the SSP
Coordinators with questions about pairs or re-pairings not
addressed in this plan.
4. Recommendations for breeding birds must await determination
of sex. Jean Dubach at Loyola
Medical Center is willing to use DNA probes to determine the sex
of any African penguin, at a low
cost to the SSP zoos. A submission form with detailed
instructions for this test is attached (Appendix
I).
5. The SSP wants to remind all institutions to check the
ownership of any birds that are recommended
to transfer. It is the responsibility of the holding institution
to inform the owner about the transfer.
6. The SSP considers all individuals, of all participating
institutions, to be a part of the SSP
population. Therefore, unless recommended by the current
Breeding and Transfer Plan, any
and all transactions must be pre-approved by the SSP Steering
Committee, including the
transfer of offspring produced as a recommendation of the
current plan. Individuals that are
not recommended for breeding should not be bred, and no
offspring are to be produced
outside of these breeding recommendations. Consult the SSP
Coordinators if you have any
questions on this matter.
7. Institutions interested in creating a new penguin enclosure
or expanding their current one are
encouraged to explore the possibility of holding Humboldt
penguins rather than African penguins. If
you are interested, contact the Humboldt penguin SSP

Explanation of Recommendations Using MateRx
MateRx is analytical software developed jointly by the National
Zoological Park and Lincoln Park Zoo. The primary output is a
matrix of genetic ratings (Mate Suitability Indices = MSI) for
every possible breeding pair in a population. MSIs allow managers
to quickly discover how the genetic status of animals in their
collections and compare to the rest of a managed population. Each
MSI represents the genetic consequences for the population if a
given pair was to produce offspring. There are seven values for
MSIs: offspring of pairs rated 1, 2, or 3 would benefit the
population’s genetic situation; pairs rated 4, 5, or 6 would be
detrimental to the population’s genetic situation. Pairs without an
MSI value (i.e., a dash [--]) should not be considered under any
circumstances without consulting a PMC, PMC Adjunct, or SPMAG
advisor. These MSI values are defined as:
1 – very beneficial genetically (recommended to double clutch) 2
– moderately beneficial (recommended to double clutch) 3 – slightly
beneficial (recommended to double clutch) 4 – neutral 5 –
moderately detrimental 6 – very detrimental genetically
MateRx integrates four genetic factors to produce the Mate
Suitability Index (MSI). These four components are used by PMC, PMC
Adjunct, or SPMAG advisors to develop breeding recommendations. In
decreasing order of “importance,” the components used to rate
pairings are: 1. the expected change in genetic diversity
(increase, decrease) that would result if an offspring of a
pair is added to the population; 2. the relative rareness or
commonness of the parents genetic information (i.e., the
difference
between the male and female mean kinships); 3. the inbreeding
coefficient of offspring that would be produced by a pair; and 4.
the proportion, if any, of the dam and/or sire’s pedigree that is
of unknown origin.
