The remaining 0.1 % matters for phenotype
What does mean this 0.1% difference ?
This translates into 3 million separate “spelling” differences (polymorphisms) in our genome book
Some/many of these differences predict inter individual variations
a) in disease susceptibility
b) in Clinical expression
c) in drug response

• Cost of Drug Development : $ 500 – 700 million for
each drug .. 80% fails in clinical trial

Pharmacogénétique : défauts moléculaires
• Les mêmes que dans les maladies génétiques : mutations ponctuelles, délétions (insertions), reéarragements géniques
• polymorphisme de l'induction ?

• dans une maladie génétique, la mutation s'exprime en fonction de son mode spécifique de transmission et produit un phénotype anormal
• en pharmacogénétique, l'expression de la mutation est conditionnelle à l'étatde base, le phénotype est normal et un phénotype anormal n'apparaît qu'en présence d'un agent extérieur xénobiotique - médicament (ex. déficit en G6PD)
