The Rspo proteins can interact with the Frizzled/LRP6 receptor complex in a manner that causes the stabilization and resulting accumulation of the intracellular signaling protein, β-catenin. This activity effectively activates and increases the subsequent nuclear signaling of β-catenin. R-Spondin can also bind to the previously discovered G-protein coupled receptors, LGR-4 and LGR-5. Rspo/β-catenin signaling can act as an inducer of the transformed phenotype, and can also regulate the proliferation and differentiation of certain stem cell populations. Recombinant CHO cell derived R-Spondins are glycoproteins containing between 222 and 251 amino acid residues. Human EGFL7 (Epidermal Growth Factor Like Protein 7)EGFL7 (Epidermal Growth Factor Like Protein 7, Multiple EGF Like Domains Protein 7, VE Statin) isa multi-domain protein containing two EGF like domains and one EMI domain. It is expressed almostexclusively in endothelial cells and functions to promote normal development of the vascular system,particularly tubulogenesis. EGFL7 is capable of antagonistic binding to Notch receptors resulting inthe inhibition of Notch signaling in HUVEC and neural stem cells. In research models inducing hypoxia and subsequent reoxygenation, (H/R), EGFL7 can inhibit ICAM-1 expression and enhancethe inhibition of NF-kappaB activation. Additionally, EGFL7 can chemoattract endothelial cells andbind to the extracellular matrix. The overexpression of EGFL7 is observed in various cancers, and isgenerally correlated with increased metastasis and a poor prognosis.

Human ANGPTL-3, ANGPTL-6ANGPTL-3 (Angiopoietin like protein 3) and ANGPTL-6 (Angiopoietin like protein 6) are membersof the angiopoietin family of structurally related proteins, characterized by a coiled N-terminal domain and a C-terminal fibrinogen like domain. These proteins are primarily expressed in the liver and can exert activities related to both angiogenesis and metabolism. ANGPTL-3 inhibitslipoprotein lipase (LPL) and endothelial lipase (EL), which has the effect of increasing plasma levelsof triglycerides and HDL associated cholesterol. The fibrinogen like portion of the ANGPTL-3 proteincan bind alpha-5/beta-3 integrins leading to endothelial cell adhesion and migration. ANGPTL-6participates in regulating glucose homeostasis by inhibiting transcriptional activity of Fox01 and reducing the production of glucose-6 phosphatase (G6Pase). Human C1inh (C1 inhibitor)C1 Inhibitor (C1inh, C1 Esterase Protein, Serpin G1) is a member of the serpin family of structurally related proteins, and is theprimary regulator of the immune complement system. C1inh is a protease inhibitor that functions to inhibit the complementsystem in order to prevent over-activation or spontaneous activation. Inhibition is achieved by binding to and irreversibly inhibiting the C1r and C1s proteases of the C1 complex, which has the effect of shutting down all subsequent downstreamevents in the complement activation cascade. C1inh can also inhibit various other proteases, including Kallikrein, Factor XIa,and Factor XIIa. Deficiencies in C1inh are the primary cause of hereditary angioedema (HAE, hereditary angioneurotic edema),a disease characterized by edema in the respiratory and gastrointestinal tracts. In certain clinical situations, the direct administration of C1inh can be used to treat HAE and certain other conditions.
