INTRODUCTION

ELISA is a trustworthy mehtod for antigen or antibody presence

evaluation.

ELISA Constitutes an evidence for immunological interactions

SEROLOGICAL EVALUATION OF IMMUNITY TO THE

VARICELLA ZOSTER VIRUS BASED ON A NOVEL

COMPETITIVEFebruary 6, 2016.

Page 6: Molecular Biology Techniques (ELISA)

VZV express 9 proteins and many of these are

immunogenic.

VZV( Varicella Zoster Virus)

Page 7: Molecular Biology Techniques (ELISA)

Investigation made possible the

identification of specific mAb for

each virus protein.*

Page 8: Molecular Biology Techniques (ELISA)

1. Detection mode: Addition of antibodies to ELISA plates to capture recombinant virus protein

2. Use of labelled neutralized mAbs to block serum antibodies

3. Analysis. Blocking rate indicates neutralizing antibody on serum

Page 9: Molecular Biology Techniques (ELISA)

I think it is a great begininig because the results of these techniques will allow the development of strategies that improve treatment of viral infections, immunization ways and identification of most expressed virus proteins and which antibodies are responsible to protect us against patogens.

Thanks to the studies realized was established a

new alternative method that open the door for new

investigations about the interactions between

monoclonal antibodies and virus proteins

IMMUNODIAGNOSIS OF FASCIOLA GIGANTICA INFECTION USING

MONOCLONAL ANTIBODY-BASED SANDWICH ELISA AND

IMMUNOCHROMATOGRAPHIC ASSAY FOR DETECTION OF CIRCULATING

CATHEPSIN L1 PROTEASE

Faculty of science, Mahidol UniversityJanuary 1, 2016.

Parasitological diagnosis of fasciolosis is possesses low sensitivity.

Detection of circulating parasite antigens is thought to be a better alternative for

diagnosis of fasciolosis.

Page 12: Molecular Biology Techniques (ELISA)

WHY WAS USED ELISA METHOD?

ESPECIFITY: 100% SENSIBILITY: 95%

ACCURACY:98.6%

Page 13: Molecular Biology Techniques (ELISA)

The study was realized in cattle and mice infected with

fasciola gigantica serum.

MoAb 4E3 and biotinylated rabbit anti-recombinant CatL1

antibody were used due to their high specificities.

Page 14: Molecular Biology Techniques (ELISA)

I think that is interesting that despite of the immense quantity of efficient methods for diagnosis and treatment of different kind of diseases, scientists are constantly searching to improve it. This investigation have allowed an efficacy increment for diagnosis of an infection caused by a parasite, constituting the first step to begin its correct and fast treatment.

MEDICAL UTILITY

Page 16: Molecular Biology Techniques (ELISA)

Main purpose of both investigations was to validate an accurate method to establish the immunity state of an

individual.

Page 17: Molecular Biology Techniques (ELISA)

The Implementation of ELISA method

allows the identification of

presence of antigens in patients and the

establishment of how is functioning their immune system in

response of thereof.

Page 18: Molecular Biology Techniques (ELISA)

I think this method can be used for epidemiological investigations and the results of these studies at the same time will contribute to the development of alternatives for

primary prevention such as better immunization programs.
