Isolation of Hydrocarbon-degrading Bacteria from Guimaras Soil Samples

Soil samples from various oil-contaminated areas in Guimaras will be obtained and

stored in 50 mL conical tubes at room temperature. To isolate possible hydrocarbon-

degrading bacteria from each sample, a pinch of soil will be inoculated into 50 mL of

liquid minimal medium (for 1 L: 0.5g KH2PO4, 2.0g KNO3, 1.0g MgSO4 7H2O, 0.0012g

CaCl2•2H2O, 0.002g FeSO4•7H2O) with 50 μL of bunker oil (Petron Corporation,

Philippines), and subjected to incubation while shaking at 37ºC at 225 rpm for one

week. Once a visible change in turbidity is observed, an applicator stick will be

dipped into the suspension culture and streak plated on Luria-Burtani (LB) agar

(Pronadisa, Madrid, Spain). One streak plate will be made for each soil sample and

incubated overnight at 37°C. Isolated bacterial colonies of different colonial

morphologies will be picked and subcultured into separate LB agar slants.

Bars represent expected duration of the task

Arrows connect prerequisite tasks

Shaded bars mean finished tasks while unshaded bars are unfinished

Shading tells how complete a task is

(think percentage)
