Measles & Immunity LabSplit your table into two lab groups
Each group will need:
one 96-well plate
two plastic pipettes
Share:IgM Test IgG Test
0.1 M Hydrochloric Acid0.1 M Sodium Hydroxide
Each 96-well plate will test 48 subjects • Rows A – D are for the IgM Test (infection antibodies)• Rows E – H are for the IgG Test (memory antibodies)• Wells A1 and E1 are for the same subject
Prepping the Plates – pgs. 3 - 4
Rows E-H
Add a drop of Sodium Hydroxide
to 35 – 45 wells
Positive wells indicate IMMUNITY
The IgG Test is phenolphthalein
Rows A-D
Add a drop of Hydrochloric Acid
to 5 – 10 wells
Positive wells indicate INFECTION
IgM Test is bromothymol blue
