The tested regions are: • AZFa: sY84, sY86, DFFRY, DBY • between AZFa and AZFb: sY95 • AZFb: sY117, sY125, sY127, sY134 • AZFc: sY254, sY255. In addition, SRY, which codes for the Testis Determining Factor, and ZFX/Y, used as internal amplification control, are amplified. The analysis of Y chromosome microdeletions is an essential diagnostic tool for the research of male infertility. The long arm of the Y chromosome has been subdivided into three regions called AZFa, AZFb and AZFc which control the correct course of spermatogenesis and often carry deletions in persons with azoospermia and severe oligospermia. Alterations in one or more loci lead to a drastic reduction in number or complete loss of sperm cells. After the Y chromosome had been sequenced and with the understanding of the mechanisms causative of the microdeletions, European Guidelines for molecular diagnosis of y-chromosomal microdeletions were established (Simoni et al., 2004).
