68. 表皮ブドウ球菌特異酵素による院内感染対策

緒 言
病原細菌に起因する感染症に対し,人類はこれまで,抗生物質を主とする薬剤を多用し,一時的な感染症の予防・治療に成功した.しかし,多剤耐性菌の出現という深刻な問題を繰り返し引き起こしている.このような“人類 vs. 細菌”の戦いは未だ決着がついておらず,新たな概念に基づいた新規感染症予防法の開発が切望されている.近年我々は,実験科学と疫学に基づき,常在細菌が病原細菌のヒトへの定着を阻害するという新たな概念を世界に先駆けて証明した 1).黄色ブドウ球菌は,健康なヒトの3割の鼻腔に存在し,この定着が院内感染のリスクファクターの一つと考えられるが,鼻腔から黄色ブドウ球菌を除菌する効果的で安全な方法は見つかっていない.我々は,常在細菌である表皮ブドウ球菌が,Esp というセリンプロテアーゼを分泌することで MRSA(メチシリン耐性黄色ブドウ球菌)を含む黄色ブドウ球菌のバイオフィルム形成を阻害し除菌することを見出した.本研究では,Esp の作用機序の解明を目指し,Esp により特異的に分解されるタンパク質(黄色ブドウ球菌の細胞表層タンパク質や黄色ブドウ球菌の定着に重要な宿主タンパク質)を明らかにしたので報告する.
方法、結果および考察
1.黄色ブドウ球菌のバイオフィルムマトリクスタンパク質と細胞壁アンカー型タンパク質の単離方法の確立と同定 まず高濃度の塩(1 M NaCl)処理により,黄色ブドウ球菌のバイオフィルムマトリクス画分を菌体表面から剥離させ,遠心分離により回収した(Fig. 1).続いて,リゾスタフィン(黄色ブドウ球菌特異的溶菌酵素)処理により細胞壁画分を細胞から遊離させ,遠心分離により回収した.それぞれの画分に含まれるタンパク質を SDS-PAGE により展開後,アミノ酸シーケンス解析およびマススペクトル解析を行った.その結果,既知のバイオフィルム関連タンパク質

3.Esp の標的タンパク質の同定と機能解析 まず,プロテアーゼ活性を失った Esp 変異体(EspS235A)を調製し,メチシリン耐性黄色ブドウ球菌(MRSA)臨床分離株のバイオフィルムの破壊効果を調べた.その結果,EspS235A はバイオフィルム破壊活性を示さなかったことより,バイオフィルム破壊にはプロテアーゼ活性が重要であることが明らかとなった.次に,上記の項目1および2で得られた成果を基盤として,Esp が分解する黄色ブドウ球菌のバイオフィルムマトリクスタンパク質および細胞壁アンカータンパク質を同定した.興味深いことに,Esp はプロテアーゼ K などのタンパク質分解酵素とは異なり,いくつかのタンパク質を選択的に分解することを見出した(Fig. 3).さらに,アミノ酸シーケンス解析,マススペクトル解析および Western blotting により,黄色ブドウ球菌のバイオフィルム中で Esp によって分解されるタンパク質を調べた.その結果,Esp は Atl,β-toxin,Eap,Emp,FnBPA,Spa といったバイオフィルム形成に関与するタンパク質のみならず,Efb,IsdA,Sbi,SceD,SdrD などの宿主への定着や感染に重要なタンパク質を分解することが判明した
