Vorwort
Diese Norm enthält sicherheitstechnische Festlegungen.
Diese Norm wurde vom Normenausschuss Medizin (NAMed) im DIN Deutsches Institut für Normung e. V., Arbeitsausschuss NA 063-04-05 AA �Sterilisatoren für pharmazeutische Sterilisiergüter� erarbeitet.
DIN 58950 �Sterilisation � Dampf-Sterilisatoren für pharmazeutische Sterilisiergüter� besteht aus:

Änderungen
Gegenüber DIN 58950-3: 2003-04 wurden folgende Änderungen vorgenommen:
a) Begriffe entsprechend an DIN 58950-1 angepasst;
b) Anwendungsbereich wurde festgelegt, dass als sterilisierendes Agens feuchte Hitze verwendet wird;
c) Anforderungen an Justierung und Kalibrierung überarbeitet;
d) der Begriff �produktbezogene Validierung� wurde geändert in �Validierung spezieller Sterilisations-verfahren�
e) Anforderungen an Geräte und Hilfsmittel zum Qualifizieren der Funktion und Leistung überarbeitet;
f) Werte für Evakuierung und Druckanstieg wurden an die Werte aus dem überarbeiteten DIN 58950-2 angepasst (siehe Anhang B);
g) unter den durchzuführenden Prüfungen nach Anhang D wurden ergänzt: die Überprüfung der baulichen Voraussetzungen und Überprüfung der Betriebsmittel in Bezug auf Qualität nach DIN 58950-7;
h) Anforderungen zur Überwachung des Dichtungsdruckes im Anhang E überarbeitet;
i) unter den durchzuführenden Prüfungen nach Anhang F wurden gestrichen: Sterilisation des Belüftungs-filters und Integritätsprüfung des Belüftungsfilters nach der Sterilisation;
j) im Anhang G wurden die Prüfungen unterteilt nach Prüfungen, die in der Leerkammer und jene, die mit Beladung durchzuführen sind;
k) Prüfung der aseptischen Bedingungen in der Kammer nach der Belüftung bei VOT bzw. VMT im Anhang G wurde gestrichen;
l) Prüfung elektromagnetischer Störfelder am Aufstellort im Anhang H gestrichen;
m) der normative Anhang I über die Prüfliste im Rahmen der Validierung wurde ein informativer Anhang;
n) die Norm wurde redaktionell überarbeitet.

1 Anwendungsbereich
Diese Norm gilt für die Prüfung von Dampf-Sterilisatoren für pharmazeutische Sterilisiergüter nach DIN 58950-2.
Sie beschreibt die Prüfungen, die vom Betreiber und Hersteller vor Verwendung des Sterilisators für die Herstellung von pharmazeutischen Sterilgütern durchgeführt werden sollen.
Dampf-Sterilisatoren nach dieser Norm sind Sterilisatoren, die als sterilisierendes Agens feuchte Hitze verwenden. Dazu zählen Verfahren, die mit Sattdampf, Dampf-Luftgemischen oder Heisswasserberieselungs-verfahren arbeiten.
2 Normative Verweisungen

3 Begriffe
Für die Anwendung dieser Norm gelten die in DIN 58950-1 angegebenen Begriffe.
4 Prüfungsarten
4.1 Überprüfung der technischen Spezifikation (en: Design Specification/Qualification)
Alle in dieser Norm festgelegten Prüfungen erfordern eine vorher festgelegte Spezifikation.
Die grundlegenden Vorgaben für die mechanische Ausführung eines Sterilisators einschließlich der Rege-lungstechnik sind in DIN 58950-2 beschrieben. Sie sind gegebenenfalls anwenderspezifisch zu ergänzen.
4.2 Prüfung durch die benannte Stelle nach Druckgeräte-Richtlinie
Die durch die benannte Stelle nach Richtlinie 97/23/EG über Druckgeräte durchgeführte erstmalige Prüfung ist vom Hersteller durch Vorlage der Konformitätserklärung nachzuweisen.
4.3 Abnahmebeurteilung (en: Installation Qualification, IQ)
Bei der Abnahmebeurteilung wird die Vollständigkeit und Fertigungsgüte der Lieferung und Installation einschließlich der Grundfunktionen der Einzelkomponenten geprüft.
Die Prüfung sollte beim Hersteller beginnen und nach der Installation beim Betreiber fortgesetzt werden.
Die Prüfung der Abnahmebeurteilung ist nach den Anhängen A bis D durchzuführen. Entsprechend den Vereinbarungen über den Abnahmeort und den technischen Möglichkeiten können die Abnahmen nach den Prüflisten nach Anhang B und Anhang C beim Hersteller oder Betreiber erfolgen.
4.4 Prüfung auf Arbeitssicherheit
Die Prüfung der Schutzeinrichtungen und -funktionen auf ordnungsgemäße Funktion der erforderlichen
Arbeitsschutzeinrichtungen ist nach Anhang E durchzuführen.
Die Prüfpunkte E.1 bis E.6 sollten beim Hersteller der Anlage erfolgen. Die Prüfungen E.7 und E.9 können erst nach der Montage der Anlage beim Betreiber durchgeführt werden.
4.5 Kalibrierung/Justierung
Die Kalibrierung der verwendeten Messgeräte (Sterilisatorinstrumentierung und externe Messgeräte) für Temperatur und Druck muss auf ein Nationales Normal (PTB oder DKD) zurückzuführen sein.
Kalibrierungen von externen Messgeräten sind vor und nach einer Serie von Prüfarbeiten (Qualifizierungen) durchzuführen, um nachzuweisen, dass während der Prüfarbeiten die Messgeräte der erforderlichen Genauigkeit entsprachen.

4.6 Funktions- und Leistungsbeurteilung (en: Operational Qualification, OQ und Performance Qualification, PQ)
Die Funktions- und Leistungsbeurteilung erfordern ordnungsgemäße bauliche Gegebenheiten und Betriebsmittelversorgungen. Sie beinhaltet die Prüfung der Programme und der Temperaturverteilung im Sterilisator und in der Beladung. Zu diesem Zeitpunkt müssen die sicherheitstechnischen Prüfungen erfolgreich abgeschlossen sein.
Zusätzlich können die automatische Be- und Entladung und weitere vereinbarte Leistungen geprüft werden.
Wenn Teile der Funktions- und Leistungsbeurteilung beim Hersteller durchgeführt werden, muss die Versorgung mit Energien/Medien beim Hersteller und Betreiber äquivalent sein. Wichtige Prüfungen sollten jedoch beim Betreiber durchgeführt bzw. wiederholt werden.
Die Funktions- und Leistungsbeurteilung sind nach Anhang F und Anhang G unterteilt nach programm-unabhängigen und programmabhängigen Prüfungen durchzuführen. Die programmabhängigen Prüfungen sind für jedes Programm durchzuführen.
Die Prüfungen mit Prüfbeladungen sind im Allgemeinen 3-mal je Beladung durchzuführen, um die Reprodu-zierbarkeit nachzuweisen.
Bei der Sterilisation von Lösungen ist die minimale und maximale Beladung des Sterilisators zu prüfen. Die Auswirkungen von Teilbeladungen sind zu diskutieren. Zur Simulation der Beladung mit Lösungen sind mit Wasser gefüllte Flaschen oder Ampullen zu verwenden, die im Rahmen der Prüfungen auch mehrfach verwendet werden können.
Für die Sterilisation von Geräten ist die maximale Beladung nach den Anwendervorgaben zu prüfen.
ANMERKUNG Für die Messung der Temperaturverteilung sollten mindestens vier Temperaturfühler in der Sterilisier-kammer der Nenngröße 6 6 6 nach DIN 58950-2:2011-01 verwendet werden; bei größeren Sterilisierkammern legen Hersteller und Betreiber die Anzahl bei Auftragserteilung gemeinsam fest.
4.7 Validierung der Programmsteuerung
4.7.1 Allgemeines
Die Regelwerke der EG und USA zu �Good Manufacturing Practice (GMP)� bezeichnen alle Arten von Pro-grammsteuerungen als �Computer� oder �computergestützte Verfahren�, die nach den entsprechenden Regelwerken zu validieren sind.
Die Validierung einer Programmsteuerung sollte als Teil des gesamten Lebenszyklus eines Systems angesehen werden. Dieser Zyklus umfasst die Stadien Planung, Spezifizierung, Programmierung, Prüfung, Inbetriebnahme, Dokumentation, Betrieb, Kontrolle und Änderungen.
Vom Hersteller serienmäßig angebotene Steuerungen sind vom Hersteller zu validieren und die Unterlagen entsprechend der Bestellung dem Betreiber zur Verfügung zu stellen.
Zusätzliche Prüfungen einer serienmäßigen Steuerung bzw. einer modifizierten Steuerung sind in Anlage H aufgelistet. Die Prüfungen für kundenspezifische Steuerungen müssen zwischen Hersteller und Betreiber gesondert festgelegt werden.
4.7.2 Prüfung des Programmausdruckes
Mathematische Funktionen/Algorithmen sollten vorzugsweise durch Kontrolle des Programmausdruckes überprüft werden.

4.7.3 Modulprüfung
Bei der Modulprüfung ist die Funktion der einzelnen Module zu überprüfen. Die Prüfung wird vom Software-hersteller nach Prüfplan (Funktionsbeschreibung des Moduls) durchgeführt und die Ergebnisse werden dokumentiert. Bei der Modulprüfung können auch Rechenfunktionen simuliert und geprüft werden (z. B. F
0-Wert-Berechnung).
4.7.4 Simulation auf einem Steuerungsprüfstand
Die Funktion der Steuerung (ohne den Sterilisator) kann auf einem Simulationsprüfstand geprüft werden. Alle Ein- und Ausgänge sind durch Signalgeber und Anzeigen zu simulieren bzw. zu überprüfen. Die statischen Funktionen der Steuerung können auf diesem Prüfstand nachgestellt und geprüft werden; dynamische Prozesse (z. B. Regelverhalten) können nicht simuliert werden und müssen an der fertigen und gegebenenfalls beladenen Anlage geprüft werden. Auf dem Prüfstand ist das Zusammenspiel der einzelnen Programm-Module zu überprüfen.
4.7.5 Abnahmebeurteilung (en: Installation/Qualification, IQ)
Nach der Installation ist die Verdrahtung der Anlage (Klemmenbelegung) zu überprüfen. Dazu sind aus der Steuerung heraus die Ausgänge zu setzen und die Reaktion der Aktoren (Ventile, Motoren, Anzeigen) ist zu überprüfen, ähnlich sind auch alle Sensoren zu aktivieren und der Eingang in die Steuerung zu überprüfen.
4.7.6 Prüfungen an der fertigen Anlage (�black-box� Prüfungen)
Prüfungen an der fertigen Anlage erfassen sowohl die Anlage (den Sterilisator) als auch die Steuerung (Hard- und Software), sind aber sehr zeitaufwendig und können nicht für alle Anforderungen durchgeführt werden.
An der fertigen Anlage müssen Probeläufe die bestimmungsgemäße Funktion für alle Programme bestätigen. Zusätzlich müssen die Sicherheitsfunktionen, die meist durch eine Kombination aus computergestützter und konventioneller Steuerung bestehen, nach Anhang E geprüft werden.
4.8 Leistungsbeurteilung (en: Performance qualification, PQ) spezieller Sterilisationsverfahren
Die Leistungsbeurteilung des Verfahrens ist produkt- und firmenspezifisch und daher in der Regel Sache des Betreibers.
Unter Leistungsbeurteilung eines Sterilisationsprogramms wird die Prüfung mit Originalprodukt verstanden. Diese Prüfung ist nur erforderlich, wenn die Daten aus der Funktions- und Leistungsbeurteilung mit Wasser oder Prüfbeladung nicht mit ausreichender Genauigkeit auf das Produkt übertragen werden können.
Die Prüfung ist für jedes Programm, im Allgemeinen 3-mal je Beladung, durchzuführen, um auch die Repro-duzierbarkeit nachzuweisen.
Die Leistungsbeurteilung spezieller Sterilisationsverfahren können nach Anhang I durchgeführt werden.
4.9 Inbetriebnahme-Zertifikat
Die erfolgreich durchgeführten Prüfungen zur Inbetriebnahme sind in einem zusammenfassenden Prüfbericht
nach Anhang J zu bestätigen.
Zusätzlich muss der Betreiber die erforderliche Prüfung vor Inbetriebnahme des Sterilisators am Aufstellungsort durch die zugelassene Überwachungsstelle bzw. den Sachverständigen gemäß BetrSichV organisieren (Sachverständigenprüfung).

5 Prüfungsorte
Die Prüfungen nach 4.2 bis 4.4 können je nach Vereinbarung beim Hersteller und/oder am Aufstellort (d. h. beim Betreiber) erfolgen. Die Prüfungen nach 4.5 bis 4.8 finden in der Regel am endgültigen Aufstellungsort des Dampf-Sterilisators statt.
6 Prüfungsumfang
6.1 Prüfungsumfang für die Abnahme
Der Prüfungsumfang für die Abnahme ist vor Auftragserteilung zwischen den Vertragspartnern an Hand der Prüflisten (Anhänge A bis I) festzulegen. Nicht durchzuführende Prüfungen sind auszustreichen.
6.2 Prüfungsumfang für eine zu vereinbarende Vorabnahme beim Hersteller
Der Prüfungsumfang für eine zu vereinbarende Vorabnahme beim Hersteller (en: factory acceptance test, FAT) ist an Hand der Prüflisten (Anhänge A bis I) festzulegen.
6.3 Anzahl der Temperaturmessungen
Die Anzahl der Temperaturmessungen bei der Funktions- und Leistungsbeurteilung und der produkt-bezogenen Validierung ist zwischen den Vertragspartnern vor Auftragserteilung festzulegen.
7 Geräte und Hilfsmittel
7.1 Geräte und Hilfsmittel zum Kalibrieren
7.1.1 Allgemeines
Die Geräte und Hilfsmittel zum Kalibrieren (siehe 4.5) stellt in der Regel der Hersteller.
7.1.2 Geräte zum Kalibrieren der Temperaturmessgeräte
Folgende Geräte zum Kalibrieren der Temperaturmessgeräte sind zu verwenden:
geeignete Thermometer mit Prüfbescheinigung mit einem Messbereich entsprechend der geforderten Sollwerte und Fehlergrenzen von 0,1 °C;
ein temperaturgeregeltes Bad oder ein Metallblock-Kalibrator mit geeigneten Einsätzen für die verwen-deten Mess- und Referenzfühler mit einem Temperaturbereich von 50 °C bis 150 °C und einer Tempe-raturkonstanz von 0,1 °C.
7.1.3 Geräte zum Kalibrieren der Druckmessgeräte
Es sind geeignete Fein-Druckmessgeräte Klasse 0,2 nach DIN EN 837-1 mit Prüfbescheinigung und einem Messbereich von 1 bar bis 3 bar, und gegebenenfalls geeignete Absolut-Druckmessgeräte der Klasse 1,0 mit Prüfbescheinigung und einem Messbereich Pabs von 0 mbar bis 250 mbar, zu verwenden.
7.1.4 Geräte zum Kalibrieren der Zeitmessgeräte
Es ist eine Stoppuhr mit einer Fehlergrenze von 2 s über die Zeitspanne von 15 min zu verwenden.

7.2 Geräte und Hilfsmittel zur Funktions- und Leistungsbeurteilung
Die Geräte und Hilfsmittel zur Funktions- und Leistungsbeurteilung stellt in der Regel der Betreiber. Hierzu gehören:
Sterilisiergüter, gegebenenfalls mit Beschickungsgeräten, für jeweils eine maximale Beladung zum Ermitteln der Temperaturverteilung;
Mehrkanalschreiber der Klasse 0,25 oder vergleichbare Aufzeichnungsgeräte mit einer Auflösung von 1 mm/K, Pt 100-Fühler Klasse A nach DIN EN 60751 oder gleichwertige Sensoren;
Filterprüfgeräte zum lntegritätstest der Sterilfilter zum Belüften bzw. zum Aufbau eines Stützdrucks. Die Prüfgeräte sind entweder Bestandteil der Bestellung oder vom Betreiber bereitzustellen. Die Prüf-durchführung und Handhabung richten sich nach den Vorgaben der Filter- und Gerätelieferanten;
bei alternativer Belüftung der Kammer ohne Sterilfilter (DIN 58950-2:2011-01, 8.3.5) Sedimentations-platten oder Flüssignährmedien zur Prüfung der aseptischen Bedingungen während der Belüftung der Sterilisierkammer mittels Exposition steriler Nährmedien;
ANMERKUNG Es wird empfohlen, Sedimentationsplatten oder Behältnisse mit Flüssignährmedien nach der Europäischen Pharmakopöe zu verwenden.
Bio-Indikatoren zur Anwendung bei �Standard-Sterilisationsverfahren�. Als Prüfkeim sind Sporen von Geobacillus stearothermophilus nach der Europäischen Pharmakopöe mit einem D-Wert von mindestens 1,5 min und 106 Sporen/Träger zu verwenden.

9 Abnahme
Die Abnahme beinhaltet die Erledigung aller vereinbarten Punkte nach Anhang A bis Anhang H.
Die Leistungsbeurteilung spezieller Sterilisationsverfahren nach Prüfliste I erfordert eine gesonderte vertragliche Festlegung.
HINWEIS � Dem Anwender der Formblätter in den Anhängen A bis J ist unbeschadet der Rechte des DIN an der Gesamtheit des Dokumentes die Vervielfältigung der Formblätter aus den Anhängen A bis J gestattet.

Prüfen der Überführung in den �sicheren Zustand� bei Betätigung der Taste �Programm-Stopp�.
Stichprobenartig ist bei verschiedenen Programmen und in verschiedenen Verfahrensschritten mehrere Male die Taste �Programm-Stopp� zu betätigen.

Angaben zur Durchführung der Prüfungen
Zu E.1
a) Grundlage der Prüfung ist die Norm DIN EN 61010-2-040 (VDE 0411-2-040):2006-02, 7.2. Berührungsschutz- bzw. Verletzungsschutzmaßnahmen an bewegten Teilen sind zu prüfen (Sichtprüfung).
b) Sofern Stetigförderer oder Winden, Hub- und Zuggeräte Bestandteil der Anlage sind, ist zusätzlich die Einhaltung der Unfallverhütungsvorschriften BGV D8 zu prüfen.
Zu E.2
a) Der Dampf-Sterilisator ist mit dem Sterilisierprogramm, das bei 121 °C die längste Sterilisierzeit hat, zu betreiben;
b) es sind 2 Betriebsabläufe unmittelbar hintereinander zu fahren;
c) während des 2. Betriebsablaufes ist die Oberflächentemperatur an der Verkleidung der Verschlusstür zu messen (Messstelle Mitte der Tür).
Zu E.3.1.1
Bildung des Spaltes zwischen Kammertür und Dichtung beim Öffnungsvorgang (siehe AD-Merkblatt A5).
Der Sterilisierdruckbehälter ist mit Luft unter einen Überdruck von 0,1 bar zu setzen. Der Zentralverschluss ist zu öffnen. Für diese Prüfung ist die Druckwarneinrichtung außer Kraft zu setzen. Es sind folgende Bedingungen durch Besichtigung zu prüfen:
a) ausreichende Bildung eines Ringspaltes zwischen Kammertür und Dichtung;
b) selbsttätiger Abbau des Innendruckes auf einen Druck 0,1 bar während des Öffnungsvorganges bis zum Außereingriffkommen der Schließelemente. Bei dieser Prüfung ist der Verschluss zügig zu öffnen.
Zu E.3.1.2
Funktion der Druckwarneinrichtungen (siehe auch AD-Merkblatt A5).
Bei geschlossener Druckwarneinrichtung und einem Überdruck von max. 0,1 bar ist ein Öffnungsversuch zu unternehmen. Der Zentralverschluss darf sich nicht öffnen. An den Bedienelementen ist eine Kraft von 200 N in Öffnungsrichtung aufzubringen.
Zu E.3.1.3
Prüfung der Verriegelung des Programmstarts und der Betriebsmittelventile mit der Endlage der Türplatte:
a) die Tür ist zu schließen und die Riegel sind so weit auszufahren, dass sie 2 mm unter den Kammer-kragen reichen;
b) es wird versucht, das Sterilisierprogramm zu starten. Das Sterilisierprogramm darf nicht starten; kein Betriebsmittelventil darf öffnen.

Sterilisierdruckbehälter für die Sterilisation von Flüssigkeiten.
Der/die Messfühler für das Überwachen der Flüssigkeitstemperatur ist/sind während der Prüfung zweck-mäßigerweise außerhalb der Sterilisierkammer anzuordnen.
a) Die Sterilisierkammertür ist zu schließen;
b) die Temperatur des Messfühlers ist bei wässrigen Lösungen auf eine Temperatur von (82 2) °C zu bringen; bei Flüssigkeiten, deren Siedetemperatur unter 100 °C liegt, ist die Temperatur am Messfühler auf das 0,8fache der Siedetemperatur einzustellen;
c) das Befehlselement �Tür auf� ist zu betätigen.
Die Tür darf sich nicht öffnen. Diese Prüfung ist für die Programmphasen �Ende� und �Betriebsbereit� durch-zuführen.
Zur Sicherheit gegen Messfühlerstörung (Kurzschluss) ist die Prüfung bei einer Temperatur 5 °C vollständig zu wiederholen. (Eintauchen in Eiswasser).

Angaben zur Durchführung der Prüfungen
Zu H.1 bis H.6
Computer und andere Geräte einer Steuerung müssen in geeigneter Umgebung aufgestellt werden, damit externe Faktoren das System nicht negativ beeinflussen können. Bei besonders gefährdeten Systemen kann es erforderlich sein, zusätzliche Prüfungen, z. B. gegen Wassereinbruch, Feuer, Löschmittel und unberech-tigten Zutritt, durchzuführen.
Zu H.10 und H.11
Die Dokumentation ist ein wesentlicher Bestandteil der Validierung eines computerunterstützten Systems. Bei der Prüfung wird nicht nur die Anwesenheit der Dokumente, sondern auch deren Verwendbarkeit und Aktualität geprüft. Nicht alle der aufgeführten Dokumente müssen beim Betreiber vorliegen. Für einige Dokumente (z. B. Beschreibung des QS-Systems, Software Development Report) ist es ausreichend, sie bei einem Audit zu prüfen und den Prüfbericht aufzubewahren.
Beim Kauf einer Nutzungslizenz bleibt die gesamte Software oder Teile davon Eigentum des Herstellers. In diesen Fällen sollte die Software zusätzlich bei einem Notar hinterlegt werden, um im Falle einer Geschäftsaufgabe oder ähnlicher Umstände Zugriff auf die Software zu erhalten. Dies wird häufig für den �Source Code� angewendet.
Die Soft- und Hardware einer Steuerung ist vom Hersteller zu prüfen (validieren). Bei einer serienmäßigen Steuerung muss der Hersteller über entsprechende Prüfunterlagen verfügen, die er dem Kunden zur Verfügung stellen kann.
Die Prüfung von �nicht serienmäßigen Steuerungen� oder �modifizierten Steuerungen� ist zwischen Hersteller und Betreiber in der Bestellung festzulegen.
Zu H.11
Die IST-Daten der gespeicherten Sterilisationen müssen sich jederzeit ausdrucken lassen.
Zu H.12
Die Eingabe oder Änderung von Daten darf nur von autorisierten Personen vorgenommen werden. Bei Systemen mit Passwortschutz wird die Zugriffsberechtigung zum System durch Eingabe von richtigen und falschen Passworten geprüft.
Zu H.16 und H.17
Für eine modifizierte Steuerung muss der Anwender sicherstellen, dass die Software in Übereinstimmung mit einem Qualitätssicherungssystem erstellt wird.
Der Anwender sollte das Qualitätssicherungssystem des Herstellers durch ein Audit überprüfen oder vom Hersteller eine Bestätigung der Zertifizierung nach der DIN EN ISO 9000-Familie für Software erhalten.

Angaben zur Durchführung der Prüfungen
Zu I.3
a) Eingebaute und zusätzliche Temperaturfühler sind nach Beladungsskizze zu verteilen (innerhalb und außerhalb des Sterilisiergutes);
b) das Sterilisationsprogramm ist durchzuführen;
c) die Temperatur ist zu dokumentieren;
d) die Temperaturverteilung ist bei allen vereinbarten Beladungen zu bestimmen.
Zu I.4
a) An jeder Temperatur-Messstelle ist die Dauer der festgelegten Temperatureinwirkung zu bestimmen;
b) die Stelle mit der kürzesten Einwirkungsdauer der Sterilisationstemperatur ist zu ermitteln.
Zu I.12
Bei Dampfsterilisation mit Vakuumverfahren:
a) die gemessenen Temperaturen in der Kammer sind mit dem der Sattdampfkurve entsprechenden Druck während der Sterilisierzeit zu vergleichen;
b) die Abweichung darf max. 0,1 bar bzw. nach Spezifikation betragen.
Zu I.13
a) Die Resistenz der Mikroorganismen (D-Wert) muss mindestens 1,5 min betragen (siehe Europäische Pharmakopöe);
b) die Mikroorganismen sind an den Temperatur-Messstellen bzw. an Stellen zu exponieren, die nicht mit Temperaturfühlern erreichbar sind;
c) als Prüfkeim sind Sporen von Geobacillus stearothermophilus nach Europäische Pharmakopöe, z. B. an Papierstreifen angetrocknet (Einsatz bei Sterilisiergut Gruppe C und D nach DIN 58950-6) oder in Lösung suspendiert und z. B. in Ampullen abgefüllt (Einsatz bei Sterilisiergut Gruppe A und B nach DIN 58950-6) zu verwenden;
d) alle Sporen müssen bei der Sterilisation mit 121 °C in 15 min abgetötet werden.
Zu I.16
Die Sterilisationssicherheit (SAL) für zu sterilisierende Arzneimittel muss mindestens 10 6 betragen.

This standard includes safety specifications.
This standard has been prepared by the Working Group NA 063-04-05 AA �Sterilisatoren für pharmazeutische Sterilisiergüter� of the Normenausschuss Medizin (Medical Standards Committee, NAMed) within DIN, the German Institute for Standardization.
DIN 58950, Sterilization � Steam sterilizers for pharmaceutical products consists of the following parts:

The Annexes A to H and J are normative, Annex I is given solely for the purposes of information.
Amendments
This standard differs from DIN 58950-3:2003-04 as follows:
a) the terms and definitions were adapted in accordance with DIN 58950-1;
b) it is specified in the scope that moist heat is used as sterilizing agent;
c) the requirements for adjustment and calibration have been revised;
d) the term �product related validation� has been changed into �validation of specific sterilization procedures�;
e) the requirements for apparatus and auxiliary means used for the qualification of operation and perfor-mance have been revised;
f) requirements for evacuating and pressure increase have been adapted to the values given in the revised edition of DIN 58950-2 (see Annex B);
g) the tests to be performed given in Annex D were supplemented by: check of the structural preconditions and quality check of the operating materials in accordance with DIN 58950-7;
h) the requirements for monitoring the sealing pressure given in Annex E have been revised;
i) among the tests to be performed according Annex F the following were deleted: sterilization of the breather filter and integrity check of the breather filter after sterilization;
j) the tests given in Annex G have been subdivided so as to distinguish between tests to be carried out in the empty chamber and those to be carried out with loading;

1 Scope
This standard applies for the testing of steam sterilizers for pharmaceutical products in accordance with DIN 58950-2.
It describes the tests the operating company and manufacturer should perform before using the sterilizer for the production of sterile pharmaceutical products.
Steam sterilizers in accordance with this standard are sterilizers which use moist heat as sterilizing agent. This includes procedures using saturated steam, steam/air mixtures or hot water spray processes.

3 Terms and definitions
For the purposes of this standard, the terms and definitions given in DIN 58950-1 apply.
4 Test types
4.1 Design Specification/Qualification
All the tests specified in this standard require a pre-established specification.
The basic provisions for the mechanical construction of a sterilizer including its control system are described in DIN 58950-2. If necessary, they shall be supplemented as required by the user.
4.2 Testing by the notified body in accordance with the Pressure Equipment Directive
The initial test carried out by the notified body in accordance with Directive 97/23/EC on pressure equipment shall be proven by the manufacturer by presenting the conformity declaration.
4.3 Installation Qualification, IQ
The installation qualification comprises of checking the delivery and installation for completeness and quality which includes testing the basic functions of the individual components.
Testing should be started at the manufacturer and should, after installation, be continued by the operating company.
The installation qualification shall be checked in accordance with Annexes A to D. Corresponding to the agreements made regarding the place of acceptance and the technical possibilities, the acceptance tests may be carried out following the checklists given in Annex B and Annex C at the manufacturer or the operating company.
4.4 Test for occupational safety
Testing of the safeguards and protective features for proper functioning of the required occupational safety equipment shall be carried out in accordance with Annex E.
The test items E.1 to E.6 should be conducted at the manufacturer of the system. The tests E.7 and E.9 can be carried out only after the system has been assembled and mounted at the operating company.

4.5 Calibration/adjustment
The calibration of the instruments (sterilizer instrumentation and external measuring instruments) used for measuring the temperature and pressure shall be traceable to a national measurement standard (Physikalisch-Technische Bundesanstalt, PTB, or Deutscher Kalibrierdienst, DKD).
Calibrations of external measuring instruments shall be carried out before starting and after a series of test procedures (qualifications) in order to demonstrate that the measuring instruments were of the required accuracy throughout the test procedures.
4.6 Operational Qualification, OQ, and Performance Qualification, PQ
For the operational and performance qualifications proper structural conditions and operating materials supplies are required. It includes checking the programs and temperature distribution inside the sterilizer and within the loading. The safety tests shall have been successfully completed at this point in time.
In addition, the automatic loading and unloading as well as other agreed services may be checked.
If parts of the operational and performance qualifications are carried out at the manufacturer, then the supplies of energy/media shall be equivalent at the manufacturer�s and the operating company. Important tests should, however, be carried out or repeated, respectively, at the operating company.
The operational and performance qualifications shall be carried out in accordance with Annex F and Annex G subdivided into tests independent of or depending on the program. The program-dependent tests shall be carried out for every program.
The tests with test loadings shall generally be carried out 3 times per loading in order to demonstrate the reproducibility.
For the sterilization of solutions, the minimum and maximum loadings of the sterilizer shall be tested. The effects of partial loadings are to be discussed. For the purpose of simulating the loading with solutions, water filled bottles or ampoules shall be used that may also be reused within the context of these tests.
For the sterilization of equipment the maximum loading as given in the user specifications shall be tested.
NOTE For measuring the temperature distribution, at least four temperature probes should be placed within the sterilizer chamber with a nominal size of 6 6 6 in accordance with DIN 58950-2:2011-01; for larger sterilizer chambers, the manufacturers and operating company specify the respective number conjointly when placing the order.
4.7 Validation of the programme control
4.7.1 General
The sets of rules of the EC and the USA on �Good Manufacturing Practice (GMP)� denote all types of program controls as �computers� or �computerized processes� to be validated in accordance with the respective sets of rules.
The validation of a program control should be considered as part of the system�s full life cycle. This cycle includes the stages of planning, specification, programming, testing, commissioning, documentation, opera-tion, inspection, and changes.
Standard control systems shall be validated by the manufacturer and he shall provide the user with the documents corresponding to the order.
Additional tests for standard control systems or modified control systems, respectively, are listed in Annex H. The tests for customized controls shall be specified separately between manufacturer and customer.

4.7.2 Check of the programme listing
Mathematical functions/algorithms should preferably be checked by reviewing the program listing.
4.7.3 Module test
In the module test, the functioning of the individual modules have to be checked. The test is carried out by the software manufacturer following the respective test schedule (functional description of the module) and the results are documented. Within the context of module tests it is also possible to simulate and test calculation functions (such as the F0 value calculation).
4.7.4 Simulation on a control test rig
The functioning of the control system (without the sterilizer) can be checked on a simulation test rig. All the inputs and outputs shall be simulated or checked, respectively, by signal transmitters and displays or indicators. The static functions of the control system can be reproduced and checked on this test rig; dynamic processes (e.g. control response) cannot be simulated and are to be tested on the finished and, if appropriate, loaded system. The interaction of the individual programme modules shall be checked using the test rig.
4.7.5 Installation Qualification, IQ
After the installation, the wiring of the system (terminal connections) shall be checked. To this end, the outputs from the control system shall be set and the response of the actuators (valves, motors, displays/indicators) is to be checked; similarly, all the sensors shall be activated and the inputs fed into the control system shall be checked.
4.7.6 Tests on the finished system (Black-box tests)
Tests on the finished system include both the system (the sterilizer) and the control system (hardware and software); they are, however, very time consuming and cannot be conducted for all of the requirements.
Test runs of the finished system shall confirm for all of the programs that the whole system operates as intended. In addition, the safety features, which usually consist of a combination of computerized and conventional controls, shall be tested in accordance with Annex E.
4.8 Performance Qualification, PQ of special sterilization processes
The performance qualification of the process is specific to the product and to the company and, therefore, within the operating company�s responsibility.
The performance qualification of a sterilization program is considered as test using the original product. These tests are required only if the data from the operational and performance qualifications using water or a test loading cannot with sufficient accuracy be transferred to the product.
The test shall generally be carried out 3 times per loading for each of the programs in order to also demonstrate the reproducibility
The performance qualification of special sterilization processes may be carried out in accordance with Annex I.
4.9 Commissioning certificate
The tests for commissioning carried out successfully shall be confirmed in a summarizing test report in accordance with Annex J.
In addition, the user shall, prior to commissioning of the sterilizer, organize the test required to be conducted on-site by the notified body in accordance with BetrSichV (local regulation).

5 Test sites
Subject to agreement, the tests given in 4.2 to 4.4 may be carried out at the manufacturer and/or at the operating company�s. As a rule, the tests given in 4.5 to 4.8 take place at the final installation site of the steam sterilizer.
6 Extent of testing
6.1 Extent of testing for acceptance
The extent of testing for acceptance shall be specified between the contracting parties using the checklists (Annexes A to I) before placing the order. Tests not to be conducted shall be crossed out.
6.2 Extent of tests at the manufacturer for a pre-acceptance to be agreed upon
The extent of tests to be conducted at the manufacturer for a pre-acceptance to be agreed (Factory Acceptance Test, FAT) shall be specified using the checklists (Annexes A to I).
6.3 Number of temperature measurements
The number of temperature measurements to be taken during the operational and performance qualifications and during the product related validation shall be specified between the contracting parties before placing the order.
7 Equipment and auxiliary means
7.1 Equipment and auxiliary means for calibrating
7.1.1 General
As a rule, the equipment and auxiliary means for calibration (see 4.5) are provided by the manufacturer.
7.1.2 Equipment for calibrating the temperature measuring instruments
The following equipment shall be used for calibrating the temperature measuring instruments:
suitable thermometers supplied with test certificates and having a measuring range corresponding to the required set-points and accuracy of 0,1 °C;
a temperature-controlled bath or a metal block calibrator with suitable inserts for the measuring and reference probes used with a temperature range of 50 °C to 150 °C and a temperature constancy of 0,1 °C.
7.1.3 Equipment for calibrating the pressure gauges
Suitable precision pressure gauges of accuracy class 0,2 in accordance with DIN EN 837-1 supplied with test certificates and having a measuring range of 1 bar to 3 bar and, if necessary, suitable absolute pressure gauges of accuracy class 1,0 with test certificates and a measuring range for pabs of 0 mbar to 250 mbar shall
be used.
7.1.4 Equipment for calibrating the time keeping devices
A stopwatch with an accuracy of 2 s over a period of 15 min shall be used.

7.2 Equipment and auxiliary means for the operational and performance qualifications
As a rule, the equipment and auxiliary means to be used for the operational and performance qualifications are provided by the user. They include:
products to be sterilized with charging equipment, if necessary, for one maximum loading each in order to determine the temperature distribution;
multi-canal recorder class 0,25 or equivalent recording devices with a resolution of 1 mm/K, Pt 100 (Platinum resistance) probes of class A in accordance with DIN EN 60751 or equivalent sensors;
filter testing equipment to be used for the integrity test of the sterile filters for ventilating or generating a supporting pressure. The testing equipment is either part of the order or to be provided by the user. Usage and test procedure are determined by the provisions of the filter and equipment suppliers;
if the chamber is ventilated in an alternative manner without sterile filters (DIN 58950-2:2011-01, 8.3.5): sedimentation plates or liquid nutrient media for checking the aseptic conditions during ventilation of the sterilizer chamber by means of exposing sterile nutrient media;
NOTE It is recommended to use sedimentation plates or containers with liquid nutrient media in accordance with the European Pharmacopoeia.
biological indicators for application when using �standard sterilization processes�. Spores of Geobacillus stearothermophilus in accordance with the European Pharmacopoeia with a D value of at least 1,5 min and 106 spores/carrier shall be used as test organisms.
8 Checklists
The checklists (Annexes A to I) are intended to be used for the purposes of specification and inspection of the delivery. Usage of the checklists is shown in Table 1.

9 Acceptance
The acceptance includes the execution of all points agreed in accordance with Annex A to Annex H.
The performance qualification of special sterilization processes following Checklist I requires separate con-tractual specification.

With regard to E.1
a) Basis for the test is DIN EN 61010-2-040 (VDE 0411-2-040):2006-02, 7.2. Protective measures for moving parts against unintentional contact or injury, respectively, shall be checked (visual inspection).
b) If continuous conveyors or winches, lifting and traction equipment are part of the system, then compliance with the accident prevention prescriptions of BGV D8 (national regulation) shall also be checked.
With regard to E.2
a) The steam sterilizer shall be operated with the sterilizing program for which the sterilizing time at 121 °C is longest;
b) 2 operating cycles shall be run directly following each other;
c) during the 2nd operating cycle the surface temperature shall be measured on the closing door panelling (measuring point: centre of the door).
With regard to E.3.1.1
The gap developing between chamber door and sealing during the opening process [see AD-Merkblatt A5 (national regulation)].
The sterilizing pressure vessel shall be pressurized with air to an overpressure of 0,1 bar. The central closure is to be opened. For this test, the pressure warning system shall be disabled. The following conditions shall be checked by inspection:
a) sufficient development of an annular gap between chamber door and sealing;
b) automatic decrease of the internal pressure to less than 0,1 bar during the opening process up to the point where the closing elements are fully disengaged. For this test the closure is to be opened quickly.
With regard to E.3.1.2
Functioning of the pressure warning systems [also see AD-Merkblatt A5 (national regulation)].
With the pressure warning system closed and at an overpressure of max. 0,1 bar try opening the door. The central closure shall not open. The operating elements shall be subjected to a force of 200 N in the opening direction.

a) The door shall be closed and the bolts shall be extended so far as for them to protrude under the chamber collar by 2 mm;

With regard to E.3.2.1
Check of the interlocking between chamber pressure and opening force:
a) the chamber shall be brought to an pressure of 0,2 bar pi 0,3 bar;
b) the command element �open the door� shall be activated. This shall not lead to an activation of the door drive element.
With regard to E.3.2.2
Check of the program start and the service valves being interlocked with the final position of the door plate:
a) the door plate shall be brought into a position at a distance of 5 mm from the final position;
b) try starting the sterilizing program. The sterilizing program shall not start and none of the service valves shall open.
With regard to E.3.3.1

With regard to E.3.3.2

b) an obstacle fitted with a force measuring device (spring scale) shall be set into the path of the closing door; when making contact the door shall not exert a force 150 N onto the obstacle in its path.
With regard to E.4
Safety check for sterilizer pressure vessels with special sealing systems, e.g. systems providing the sealing effect by applying an service (inflatable seals, blow-off seals, etc.).
a) The push button for monitoring the pressure of the service used for sealing shall be disconnected;
b) the program shall be checked manually during at least two stages (e.g. heating and sterilizing);
c) for control-independent electric blocking of the service valves, the electric diagram and the execution of the wiring shall be subjected to a visual inspection.
Under no circumstance may one of the service valves leading into the interior of the sterilizer chamber be triggered and/or opened. A warning signal shall sound.

With regard to E.5
Sterilizing pressure vessels for the sterilization of fluids.
The measuring probe(s) for monitoring the fluid temperature shall, for practical reasons, be positioned outside the sterilizer chamber.
a) The sterilizer chamber door shall be closed;
b) for aqueous solutions, the temperature of the measuring probe shall be brought to (82 2) °C; for fluids boiling at less than 100 °C, the temperature shall be set to 0,8 times the boiling temperature;
c) the command element �open the door� shall be activated.
The door shall not open. This test shall be conducted for the program stages �End� and �Ready for use�.
In order to safeguard against short circuit of the measuring probes the full test shall be repeated at a temperature 5 °C (ice water immersion).
With regard to E.6
For sterilizing pressure vessels which, when used as intended, are fully or partially filled with water or other fluids and the filling level of which is above the threshold or the bottom edge of the closure.
a) The sterilizer chamber door of the steam sterilizer shall be closed;
b) the level switch for the fluid level shall either be disconnected or be brought into the position �Level exceeded�;
c) the command element �open the door� shall be activated;
d) the door shall not open.
With regard to E.7
Check of the exit route from the utility room.
Applicable only for steam sterilizers with powered movement of the sterilizer chamber door and obstruction of the exit route from the utility room:
a) with the sterilizer chamber door closed the switch-off device for the door motion inside the utility room shall be activated;
b) following this the program shall be set to �open the door�;
c) the door shall not move.

With regard to F.1, F.2 and F.3
a) The temperature probes shall be compared with verified thermometers in a temperature-controlled bath in accordance with 7.1.2; it is recommended to conduct a two-point measurement in the range of the sterili-zation temperature (100 °C to 120 °C);
b) the pressure gauge shall be compared with a verified precision pressure gauge in accordance with 7.1.3.;
c) in the event of inadmissible deviations adjustments shall be made;
d) the set-points and the actual values or their deviations, respectively, shall be documented � before and after adjustment, if necessary.

With regard to H.1 to H.6
Computers and other devices of a control system shall be installed in a suitable environment so that external factors cannot affect the system. For systems particularly at risk it can be required to carry out additional tests, e.g. with regard to water ingress, fire, extinguishing agents, and unauthorized access.
With regard to H.10 and H.11
Documentation is an essential part of the validation of a computerized system. The documentation check includes not only checking the documents for completeness but also for usability and their being up-to-date. It is not necessary for all of the listed documents to be available at the operating company. For some of the documents (e.g. description of the QA system, Software Development Report) it is sufficient to have them reviewed in an audit and to retain the corresponding audit report.
When purchasing a user license, the software in its entirety or in parts continues to be the software company�s property. In cases such as this, the software should also be deposited with a notary in order to have access to it in the event of a shut-down of business or similar situation. This is frequently applied to the �source code�.
The software and hardware of a control system shall be tested (validated) by the manufacturer. For standard systems the manufacturer shall have appropriate test documents which he can provide the customer with.
The testing of �non-standard systems� or �modified systems� shall be specified between manufacturer and operating company when placing the order.

With regard to H.11
It shall be possible, at any time, to print the actual data of the stored sterilization cycles.
With regard to H.12
Data may only be entered or modified by authorized persons. For systems with password protection, the authorization for system access is checked by entering correct and wrong passwords.
With regard to H.16 and H.17
For modified systems, the user shall ensure that the software is written in correspondence to a quality assur-ance system.
The customer should check the manufacturer�s quality assurance system by means of an audit or receive from the manufacturer a confirmation of the certification in accordance with the DIN EN ISO 9000 series for software.
With regard to H.21
It should be possible to print the parameter list from the system � at least with the manufacturer�s support.
With regard to H.22
As far as required, the system should check entry and processing of data for correctness. Critical steps and functions shall be monitored by the system. Due to the different requirements, the monitoring functions shall be specified between customer and manufacturer (e.g. report of the sterilization being completed, verification of batch and product data entries, etc.).
