Excite a large panel of exogenous or endogenous fluorophores
Image several fluorophores simultaneously even with very
distant TPA spectra
Adapt the excitation to the TPA spectra

Wideband excitation for multiphoton imaging: literature
Typically: spectral broadening of femtosecond pulses in a photonic crystal fiber
(PCF) associated with a step of pulse shaping for temporal compressing
Dong Li 2009
(Applied Physics Letters)
Interesting solution answering the
questions, BUT:
Complex technically (not turn-key solution)
Expensive
Unstable in the time

Conclusion sur les résultats
Two-photon fluorescence and second harmonic generation
Similar between a nanosecond supercontinuum source and classical Ti: Sapphire laser
Good contrast in second harmonic generation
Progress to do for increasing the two-photon fluorescence contrast adjusting better the
excitation spectrum to the two-photon absorption one
Démonstration de principe réussie
mais utilisation d’un système commercial non adapté pour
l’objectif du projet ARKENCIEL
Achat d’un microscope 2P tout ouvert
