Weigh Capsule on Torsion balance

If you using a torsion balance part the weigh ensure that thebalance is clean, level on zero

Then place an empty capsule shell of the same size you areusing to punch the capsule on the left hand pan of thebalance, and dial in the selected weight for the content ofyour capsule.

Should the weight for the content of your capsule be lessthan the selected target weight, as indicate in this slide, thenlock the torsion balance and remove the capsule from theweighing pan

Then separate cap from the body oh this capsule, punch amore powder into the body of the capsule slightly recaps thenreweight

Should the weight for the content of your capsule be morethan the selected target weight, as indicate in this slide, thenlock the torsion balance and remove the capsule from theweighing pan

Continue this process of adding and removing powder fromeach capsule into the pointer align with the sign of markingon the scale as indicated here, indicating that the targetweight has been attained

Weigh Capsule on Digital balance

If you using a electronic balance part the weight as with thetorsion balance ensure that the balance is clean and level

Then place a small weighboard and the single gelatincapsule of the same size you are using to punch the capsuleon the weighing surface

Then tare the balance

Remove the empty gelatin capsule from the weighboard andreplace with the punch capsule

Should the content of the capsule weight less than desiretarget weight, then remove the capsule from the scale.

Remove the caps of the capsule from the body, punch inmore powder, gently recap then reweight

Should the content of the capsule weight more than desiretarget weight, again remove the capsule from the scale.

Remove the caps of the capsule from the body, empty outsome of the powder from body of the capsule, gently recapthen reweight

Continue this process of adding and removing powder fromthe capsule until the desired weight is attained

Continue punching all capsules

Continue punching and weighing each individual capsule untilall of the capsule have been prepared

Inspect the final capsule for esthetic appearance and polishthem using a clean wipe if necessaryInspect the final capsule for esthetic appearance and polishthem using a clean wipe if necessary

Calculation

Since the prescription instruct to prepare 60mL of suspensionat concentration of 25mg/5mL, we can easily calculate theamount of HCT required for the prescription to be 300mg

Weigh the recommended suspending agent, 1,5 – 2%Weigh the recommended suspending agent, 1,5 – 2%

Finnaly, by selecting sodium benzoate as a preservative atconcentration 0,2%, we can calculate 1,2 g of sodiumbenzoate would be needed

Richie A. I. Chandra

Page 18: 2. CND

Mixing

Selecting to use comercially available 50 mg HCT, we canagain easily determine that 6 of 50 mg tablet would benecessary to prepare the prescription

Combine the sodium benzoate and methylcellulose with HCTpowder residing in the mortarCombine the sodium benzoate and methylcellulose with HCTpowder residing in the mortar

Richie A. I. Chandra

Page 19: 2. CND

1. Obtain HCT Tablet

a. The first step in the procedure is to obtain six 50 mg HCTtablet

b. These tablet would then added to mortarb. These tablet would then added to mortar

2. Crush tablet using trituration

The tablet then crush using trituration until a very fine powder

Richie A. I. Chandra

Page 20: 2. CND

3. Weighing suspending agent

a. The next step is to accurately weigh 120 mg ofpreservative, sodium benzoate

b. Then we must weigh the recommended suspending agent,Citrucel in this example is 2g for 60 mL of suspensionb. Then we must weigh the recommended suspending agent,Citrucel in this example is 2g for 60 mL of suspension

4. Geomtrically mix powder ingredients

Next combine the sodium benzoat and citrucel with the crushHCT powder residing the mortar, mix until uniform, homogenous.

Richie A. I. Chandra

Page 21: 2. CND

5. Form primary suspension

In this point, gradually add approximately 40 mL of purifiedwater to the mixture

6. Transfer the suspension to the bottle

Now transfer these primary suspension into 60mLprescription bottle

Richie A. I. Chandra

Page 22: 2. CND

7, Rinse mortar with purified water

a. Using a small amount of purified water, rinse the mortarthroughly with channel trituration in order to retrieve anyresidue product remaining in the mortar

a. Using a small amount of purified water, rinse the mortarthroughly with channel trituration in order to retrieve anyresidue product remaining in the mortar

b. Then transfer the rinsing to the prescription bottle

c. Use purified water to qs the product to the 60 mL mark onthe prescription bottle

8. Cap bottle and shake vigorously

a. Cap the prescription bottle seecurely

b. Shake vigorously to ensure that the suspension iscompletely mixedb. Shake vigorously to ensure that the suspension iscompletely mixed
