Human epidermal growth factor receptor-2 (HER2/erbB-2) belongs to the EGF-R (HER) family of transmembrane receptors involved in signal transduction pathways that regulate cell growth and differentiation.
HER1 binds to several ligands: epidermal growth factor (EGF),transforming growth factor alpha (TGF), amphiregulin (AR), heparin-binding EGF-like growth factor (HB-EGF), betacellulin and epiregulin. HER2 has no known ligand. HER3 and HER4 bind to many neuregulins or neu differentiation factors (NDFs), a family of structurally diverse peptides.All HER ligands have a 50-55 AA EGF-like motif.
No HER2-specific ligand has been identified, but HER2 is the preferred heterodimerization partner for other HER receptors. There is a clear bias toward heterodimers containing HER2!! These heterodimers are more potent.
HER3 has no kinase activity and is the major and most potent dimerization partner of HER2.HER3 homodimers are mitogenically inactive.
However, HER3 has many docking sites for signaling molecules.
HER1(kinase)-HER3(signalling domain) chimeric receptor displays greater mitogenic effect than the HER2/HER3 heterodimer and HER1 alone.HER3’s very low kinase activity may be a method of regulation!!
HER receptors exist as monomers but dimerize on ligand binding. Interactions within the HER family do not occur at random.
Heterodimers containing HER2 generate more potent signals
Why?
Heterodimers are characterized by a relatively slow rate of ligand dissociation.
HER2 undergoes a slower rate of ligand-induced endocytosis compared with other HER receptors.
Therefore, heterodimers containing HER2 have a particularly high signalling potency compared with non-HER2 containing dimers.
Taken together, signaling by HER2 heterodimers is relativelyprolonged and results in enhanced activation of signaling pathways such as the MAPK/Akt route.
Ligands bound to the receptor are endocytosed and these vesicles form the early endosome.There, the ligand is separated from the receptor and the receptor either recycles to the cell surface or is transported with the ligand to the lysosome, where it is degraded. The HER1 homodimer is mostly degraded.
However, when HER1 heterodimerizes with HER2, it is returned to the cell surface by HER2. This recycling prolongs signaling.

The antitumor efficacy of HER2-specific antibodies such as Herceptin® relates to their ability to direct HER2 to a Cbl- dependent endocytosis and degradation pathway.
Unlike HER1, which stronglybinds c-Cbl, HER2 only weaklyinteracts with it.
The ring finger domain of c-Cbl serves as a binding site for an ubiquitin-processing enzyme (E2).
Anti-tumor antibodies tag HER2 for degradation by recruiting c-Cbl and inducing ubiquitination of HER2.
What are the possible effects of antibodies targeting a receptor?Will every antibody induce degradation of the receptor?
- They can bind to the receptor but have no effect on either its function or its half-life
- Bind to the ligand-binding site, dimerize and induce a conformational change in the receptor
- Bind to a dimerization interface and prevent dimer formation
- Cause a conformational change that enhances internalization and ubiquitination, without activating the receptor (like Herceptin®)
