OBJETIVE
• Evaluate and describe the utility of virulence gene determination as a complementary tool for epidemiological typing of MRSA in relation to spa typing and pulsed-field gel electrophoresis (PFGE).

AISLAMIENTOS
• Almacenados los MRSA con genotipo PFGE desde 1983 en Suecia.
• Determinación de patrones visuales (TENOVER – BioNumerics 6.6)

• Registro de datos para cada aislamiento.
• Descripción de los aislamientos.
• Fueron escogidos 63 aislamientos entre 1994-201, que representaban los MRSA más frecuentes de área.

• Emplear ciclos de altas y bajas temperaturas alternadas para separar las hebras de ADN recién formadas entre sí tras cada fase de replicación.
• Dejar que las hebras de ADN vuelvan a unirse para poder duplicarlas nuevamente.

METODOLOGÍA
• Desnaturalización del DNA para dar hebras sencillas
• Hibridación especifica de la hebra sencilla con un oligonucleótido
• Replicación de la hebra sencilla por una DNA polimerasa a partir de oligonucleótidos anterior como cebador.

Comparación entre las cepas para el propósitos epidemiológicos
Se identificaron 30 genes relacionados con la virulencia
9 genes de adhesinas 21 genes tóxicos
Relación de los factores de virulencia con los genes

Carecieron de los genes lukm, seh, etb, sed, selr, eta, sea, selp, seb y etv.
Presentaron los genes Ipb, cIfA, cifB y fnbB
Todos las S. Aureus aislados

ST8 (VF A) recogido en 7

ST30 (VF C) recogido en 16

La elección del método de tipificación molecular depende del propósito de la investigación
Los resultados demuestran claramente que ninguno de los métodos de genotipificación pueden identificar por si solos la tipificación epidemiológica

When conducting research must keep of the comparison of strains with epidemiological and clinical purposes, to maintei infection control.
With each research project must respect the established protocols and make a proper selction of molecular quantification methods for good typing
