Nonclinical Pharmacokinetics and Product Metabolism

• A computational analysis of COA Mixture® (COA FS®) revealed that some PIs that are currently in use showed inhibitory activities on CYP3A4.

• Seven of the phytochemical compounds present in the COA Mixture® were also predicted to possess inhibitory activity on CYP3A4.

• The inhibition of CYP3A4 by the phytochemical compound could decrease the elimination and pumping out of the four PI drugs from the systemic circulation and the cells respectively thereby increasing the effectiveness of the PI drugs (Kehinde et al., 2019).

The result also showed the drug likeness of four FDA-approved ARV drugs and fifteen

selected phytochemical compounds from COA Mixture®. Two of the four conventional

drugs and nine of the phyto-chemical compounds passed the drug likeness test. These

showed that the nine phytochemical compounds have good drug properties as seen in the

two conventional drugs (Kehinde et al., 2019).
